摘 要:目的:检测上皮性卵巢癌中Y-盒结合蛋白1(YB-1)和黏附分子CD44的表达,分析其与卵巢癌化疗耐药的关系。方法:采用免疫组化SP法分别检测YB-1及CD44蛋白在铂类耐药或敏感的上皮性卵巢癌及正常卵巢组织中的表达情况,实时荧光定量PCR(qRT-PCR)、Western blot法分别检测上皮性卵巢癌顺铂敏感与耐药细胞中YB1及CD44 mRNA与蛋白水平,分析两者表达的相关性。结果:YB-1和CD44在上皮性卵巢癌组织中阳性率分别为56.3%和60.9%,耐药组织中表达均明显高于敏感组织,且两者表达呈正相关(r=0.456);顺铂耐药上皮性卵巢癌细胞株中YB-1和CD44 mRNA及蛋白水平明显高于敏感株(P均<0.05)。结论:YB-1和CD44与上皮性卵巢癌铂类耐药相关,独立或联合检测可能预测化疗的敏感性。Objective: To investigate the expressions of Y-box binding protein 1(YB-1)and CD44 in the epithelial ovarian cancer and analyze the relationship between their expressions and ovarian cancer chemotherapy resistance.Methods: Immunohistochemical SP method were used to detect the expressions of YB-1 and CD44 protein in the tissue from platinum-resistant or sensitive epithelial ovarian cancer patient and 10 cases of normal ovarian tissue,real-time quantitative PCR(qRT-PCR),Western blot were used to detect YB1 and CD44 mRNA and protein levels in platinum-sensitive and resistant epithelial ovarian cancer cells,a comprehensive analysis was conducted of the correlation between the expression of YB1 and CD44 and the relationship between their expression and ovarian cancer drug resistance.Results: Expressions of YB-1 and CD44 in epithelial ovarian cancer tissues were significantly higher than normal ovarian tissue,the positive rates in the platinum-resistant tissue were 56.3%,60.9% respectively,which were significantly higher than that in the platinum-sensitive tissue.Moreover,their expressions were positively correlated(r = 0.456).The mRNA and protein levels of CD44 and YB-1 in platinum-resistant ovarian cancer cell lines were significantly higher than that in the platinum-sensitive ovarian cancer cell lines(P〈0.05).Conclusion: YB-1 and CD44 were associated with platinum-resistant in the epithelial ovarian cancer,and the independent or joint detection may predict chemotherapy response of epithelial ovarian cancer.
