摘 要:目的:研究透骨香叶中槲皮素的最佳提取工艺。方法:以透骨香叶槲皮素得率为指标,用反相高效液相色谱法测定,通过单因素试验,再采用正交试验设计L9(34)优化实验条件,并且研究对比了超声-机械振荡辅助提取法与常规加热回流和超声辅助提取透骨香叶中槲皮素的得率。结果:确定了超声-机械振荡辅助提取透骨香叶中槲皮素的最佳工艺为以乙醇-6.5mol/L HCl(4∶1)混合液为提取剂,料液比1∶70(g/mL),在水浴温度为65℃下超声提取40min后,再室温振荡萃取30min。优化的超声-机械振荡提取透骨香叶中槲皮素提取率分别比常规加热回流和超声辅助提取法提高了47.1%和44.9%。结论:该方法可以简便、高效地从透骨香叶中提取槲皮素,为透骨香叶中槲皮素的有效开发和利用提供一定参考依据。Object:The aim is to study the quercetin extraction from the leave of Gaultheria leucocarpa.Methods:The yield of quercetin is as the index,which is detected by ultrasonicmechanical oscillation assisted extraction(UMOAE)and heating reflux extraction and ultrasonic assisted extraction.RP-HPLC is used to determine the content of quercetin in extracts,the single factor experiment and orthogonal experimental design L9(34)are used to optimize the conditions of UMOAE.Results:The optimal process of UMOAE of quercetin from the leave of Gaultheria leucocarpais decided,that is ethanol-6.5mol/L HCl(4∶1)mixture solution as extraction solvent,the ratio of raw material to solvent of 1∶70(g/mL),immersion time of 3h,ultrasonic extraction for40 min in water bath at 65 ℃and mechanical oscillation assisted extraction for 30 min in water bath at room temperature.The yield of quercetin by UMOAE is 47.1% higher than that by heating reflux extraction and 44.9% higher than that by ultrasonic assisted extraction respectively.Conclusion:The method is simple and efficient,so as to give a reference for efficient development and utilization of quercetin in the leaves of Gaultheria leucocarpa.
