当归提取物对S180荷瘤模型小鼠肉瘤的抑瘤作用的实验研究

摘　　要：目的:探讨当归醇粗提物的抗肿瘤作用与机制。方法:昆明种小鼠60只接种腹水瘤,建立S180荷瘤模型小鼠,当归醇粗提物100、200、400 mg/kg剂量灌胃给药2周,计算模型小鼠的抑瘤率,脾指数和胸腺指数;MTT法测定当归水提物对模型小鼠脾淋巴细胞增殖的影响;ELISA法测定模型小鼠血清中肿瘤坏死因子(TNF-α)及白介素(IL-2)表达。结果:当归醇粗提物可显著抑制S180荷瘤模型小鼠实体瘤的生长(P<0.05),提高脾和胸腺指数(P>0.05),促进Con A诱导的脾淋巴细胞增长和血清中TNF-α及IL-2的表达。结论:当归醇粗提物对S180实体瘤模型小鼠有抑制肿瘤生长作用,其作用机制可能与其具有促进免疫功能有关。Objective: To explore antitumor effects and mechanism of crude ethanol extracts of Angelica sinensis. Methods:The healthy Kunming mice were inoculated with ascites tumor,and then S180 tumor-bearing mice model was established,The dose at100,200 and 400 mg·kg-1 of the extracts of Angelica sinensis were administrated for 2 weeks to determine the tumor-bearing mice tumor inhibition rate,thymus index and spleen index. MTT assay was used to detect the effects of the extracts of Angelica sinensis on the spleen lymphocyte proliferation of the tumor-bearing mice,and ELISA was used to determinate of the expression of tumor necrosis factor( TNF-α) and interleukin( IL-2) in the serum of the tumor-bearing mice. Results: The extracts of Angelica sinensis could significantly inhibited the growth of S180 solid tumor( P < 0. 05),improve the spleen index( P < 0. 05),improve the thymus index and spleen index( P > 0. 05),increase the proliferation of spleen cell by Con A induction and improve the levels of the tumor-bearing mice serum TNF – α and IL-2. Conclusions: The extracts of Angelica sinensis have inhibiting effect on S180 solid tumor,and the mechanism is related to its immune regulation function.
