摘 要:目的:观察姜黄素联合紫杉醇对乳腺癌MDA-MB-231细胞迁移侵袭能力的抑制作用及对MMP9体内外表达的影响。方法:采用细胞划痕实验检测姜黄素联合紫杉醇对MDA-MB-231细胞迁移的影响;采用Western blotting检测MDA-MB-231细胞和MDA-MB-231细胞裸鼠移植瘤组织中MMP9蛋白的表达;实时荧光定量PCR检测MMP9 mRNA的表达。结果:在体外细胞实验中,姜黄素10μmol/L组、紫杉醇0.001μmol/L组可分别降低MDA-MB-231细胞的迁移能力,并可降低MMP9蛋白和mRNA的表达,10μmol/L姜黄素和0.0005μmol/L紫杉醇联合药物组可协同减弱细胞的迁移能力并降低MMP9的表达。裸鼠移植瘤模型实验中,姜黄素100mg/kg组、紫杉醇10mg/kg组,可分别降低MMP9蛋白和mRNA的表达,姜黄素100 mg/kg与紫杉醇5mg/kg联合组可协同降低其表达。结论:姜黄素联合紫杉醇在体内外可协同降低乳腺癌MDA-MB-231细胞迁移侵袭能力。Objective: To observe the inhibitory effect of curcumin combined with paclitaxel on breast cancer MDA-MB-231 cells and the effect on matrix metalloproteinase 9( MMP9) expressions in vivo and in vitro. Methods: Using Scratch test to observe the capacity of migration in breast cancer cell,western blotting was applied to examine the protein of MMP9 in cell and xenografted tissues,Real-time quantitative PCR was used to detect MMP9 mRNA. Results: In vitro experiments,curcumin 10μmol / L group,paclitaxel 0. 001μmol / L group,reduce the migratory ability of MDA-MB-231 cells respectively,and reduce the protein and mRNA expression of MMP9,10μmol / L curcumin combined with 0. 0005μmol / L paclitaxel group synergistically decreased cell migration and reduce the expression of MMP9. Xenograft tumor model experiments,curcumin 100 mg / kg group,paclitaxel 10 mg / kg groups,reduce protein and mRNA expression of MMP9 respectively. curcumin100 mg / kg combined with paclitaxel 5mg / kg group reduce its expression synergistically. Conclusion: Curcumin combined with paclitaxel in vivo and in vitro synergistically to reduce breast cancer MDA-MB-231 cell invasive ability.
