摘  要:目的:研究葛根提取物对糖尿病视网膜病变大鼠血清氨基酸代谢的影响。方法:高脂饲料喂养6周后,腹腔注射链脲佐菌素(streptozotocin,STZ)40 mg/kg建立糖尿病视网膜病变大鼠模型,造模成功后随机分为模型组,羟苯磺酸钙组(150 mg/kg),葛根提取物组(421 mg/kg),另设正常对照组。动态监测体重及血糖变化,灌胃给药10周后取血清采用AccQ·Flour试剂进行柱前衍生化,AccQ·Tag法专用C18氨基酸分析柱进行分离,高效液相色谱荧光法(λex=250 nm,λem=395 nm)进行氨基酸含量测定。结果:葛根提取物(421 mg/kg)对糖尿病视网膜病变模型大鼠体重、血糖均无明显影响。在建立的色谱条件下,17种氨基酸的浓度与峰面积线性关系良好,日内及日间精密度RSD分别<9.28%,8.72%,平均回收率>75.02%。与正常对照组比较,模型组血清支链氨基酸(亮氨酸、异亮氨酸、缬氨酸)、酪氨酸、精氨酸、谷氨酸含量明显升高。与模型组比较,葛根提取物(421 mg/kg)干预后亮氨酸及谷氨酸浓度显著降低,异亮氨酸及酪氨酸浓度有降低趋势。结论:支链氨基酸水平升高是糖尿病视网膜病变发生的重要指征,葛根提取物对糖尿病视网膜病变大鼠的治疗作用可能与其改善氨基酸代谢紊乱有关。Objective: To study the effect of Pueraria lobata( Willd.) Ohwi extract on serum amino acid metabolism in diabetic retinopathy rats.Methods: The diabetic retinopathy model was established by intraperitoneal injection of streptozotocin( 40 mg / kg) combined with a high fat diet. The model rats were randomly divided into model control group,positive control group and Pueraria lobata( Willd.) Ohwi group( 421 mg / kg). Other ten healthy rats were regarded as the normal control group. After treatment for 10 weeks,the serum was derivatized by Acc Q·Fluor reagent and separated by Acc Q·Tag dedicated C18 column. The concentration of amino acid was detected by a fluorescence detector( λ ex = 250 nm,λ em = 395 nm). Results: Pueraria lobata( Willd.) Ohwi extract( 421 mg/kg) has no significant effect on body weight and blood glucose of rats. The calibration curves showed linearity over the studied concentration range. The within and between day variation coefficients were lower than 9. 28% and 8. 72%,respectively. The spiked recoveries were greater than 75. 02%. Compared with the normal group,the content of branched chain amino acid( BCAA,leucine,isoleucine and valine),tyrosine,arginine and glutamate were significantly increased in model group rats( P 0. 01). Treatment with Pueraria lobata( Willd.) Ohwi( 421 mg / kg) can obviously decrease the leucine and glutamate levels( P 0. 05). Conclusion: The elevation of the BCAA level can be regarded as an important indicator for the occurrence of diabetic retinopathy. The efficacy of Pueraria lobata( Willd.) Ohwi on diabetic retinopathy rats may be associated with the improvement of the metabolic disorder of amino acid.
