摘 要:目的探讨散发性乳腺癌患者乳腺癌易感基因1(BRCA1)mRNA与蛋白的表达变化及差异。方法应用荧光定量聚合酶链反应(FQ—PCR)检测正常乳腺组织(n=40)与散发性乳腺癌组织(n=80)中BRCA1mRNA的表达量变化;用免疫组织化学方法检测BRCA1蛋白表达变化及定性观察BRCA1蛋白亚细胞定位的变化,分析BRCA1与散发性乳腺癌发生发展的相关性。结果(1)散发性乳腺癌组织中BRCA1mRNA表达水平(0.759±0.440)低于正常乳腺组织(1.530±0.810),差异有统计学意义(P〈0.05),原位癌伴微小浸润组(0.956±0.340)与浸润癌组BRCA1(0.561±0.443)表达差异有统计学意义(P〈0.01),原位癌组表达水平高于浸润性乳腺癌组。(2)BRCA1蛋白在正常乳腺组织高表达,在乳腺癌组织中表达降低,其阳性率分别为95%和46%,差异有统计学意义(,=27.040,P〈0.01);浸润性乳腺癌组织中BRCA1蛋白表达水平低于导管原位癌伴微小浸润组织,差异有统计学意义(x^2=4.037,P〈0.05)。本研究还观察到BRCA1蛋白在正常乳腺组织中表达于细胞核,在乳腺癌组织中可以见到BRCA1蛋白在细胞核与细胞质中均表达,且在细胞核上的表达率降低。结论BRCA1mRNA与蛋白在散发性乳腺癌疾病进展过程中表达变化存在差异。Objective To investigate the changes in mRNA and protein expression of breast canc- er susceptibility gene 1 ( BRCA1 ) during the development and progression of sporadic breast cancer. Meth- ods BRCA1 mRNA expression in sporadic breast cancer tissues ( n = 80) and normal breast tissues ( n = 40) was detected by real - time fluorescent quantitative polymerase chain reaction ( FQ - PCR). The BRCAI protein expression and subcellular location were determined by immunohistochemistry. The correla- tion between BRCA1 and development and progression of sporadic breast cancer was analyzed. Results ( 1 ) BRCA1 mRNA expression was lower in breast cancer tissues (0. 759 ± 0. 440 ) than in normal breast tissues (1. 530 ± 0. 810), and significantly higher in carcinoma in situ (with minimal invasion) (0. 956 ± 0. 340) than in invasive breast cancers (0. 561 ± 0. 443, P 〈 0.01 ). (2) BRCA1 protein was highly ex- pressed in normal breast tissues, and lowed expressed in breast cancer tissues (positive rate: 95% vs. 46% ,x^2 = 27. 040 ,P 〈 0. 01 ). BRCA1 expression was also significantly lower in invasive breast cancer tis- sues than in ductal carcinoma in situ (with minimal invasion) (x^2 =4. 037 ,P 〈0. 05). We also observed alterations in subcellular location of BRCA1 protein confined in the nucleus in normal breast tissues, and diffused in both nucleus ( at decreased level) and the cytoplasm in breast cancer tissues. Conclusion BRCA1 mRNA and protein expression altered with the development and progression of sporadic breast cancer.
