川陈皮素通过下调磷酸肌醇3激酶/蛋白激酶B信号通路诱导凋亡抑制人乳腺癌细胞MCF-7增殖

摘  要:目的体外观察川陈皮素对乳腺癌细胞增殖的抑制作用并探讨机制。方法采用噻唑蓝(MTT)法观察川陈皮素对人乳腺癌细胞MCF-7增殖影响;Hoechst33342染色观察川陈皮素处理后细胞形态的变化;膜联蛋白V(AnnexinV)/碘化丙锭(PI)检测川陈皮素对MCF-7细胞凋亡的影响;Westernblot检测川陈皮素处理后MCF-7细胞中蛋白表达的改变。结果10μmol/L川陈皮素能明显的抑制MCF-7细胞生长(P〈0.05),抑制作用呈剂量、时间依赖关系;10、20、40μmoL/L川陈皮素处理24h后,MCF-7细胞出现细胞核皱缩,凋亡小体,并且细胞凋亡率(13.5%、37.7%、49.96%)随着川陈皮素作用浓度的增加而上升;川陈皮素能引起B细胞淋巴瘤/白血病-2相关X蛋白(bax)/B细胞淋巴瘤/白血病-2(bcl-2)比率上升(P〈0.01),磷酸肌醇3激酶(P13K)/蛋白激酶B(Akt)信号通路相关蛋白表达下降(P〈0.01)。结论川陈皮素在体外能抑制乳腺癌细胞增殖,其作用机制可能与抑制P13K/Akt信号通路从而诱导细胞凋亡有关。Objective To investigate whether nobiletin suppresses the cell proliferation of human breast cancer cell and explore the underlying mechanism. Methods Methyl thiazol tetrazolium (MTT) as- say, Hoechst 33342 stainning and Annexin V/propidium iodide (P1) were used to examine the viability and apoptosis of MCF -7 cells treated with nobiletin. Results Nobiletin of 10 μmol/L significant inhibited MCF - 7 cell proliferation ( P 〈 0.05 ) , the inhibition was revelant to dosage and time - dependant manner. After 24 hours incubated with Nobiletin of 10, 20, 40 μmol/L respectively, MCF -7 cells became shrink- age and induced apoptosis, the apoptosis rate ( 13.5% , 37.7% , 49.96% ) increased as the increasing dosage. The ratio of B cell lymphoma/leukemia - 2 ( bcl - 2 ) associated X protein/bcl - 2 was increased whereas the proteins expression of phosphatidylinositol 3 kinase/protein kinase B (PI3K/Akt) signaling pathway was decreased ( P 〈 0. 01 ). Conclusion Nobiletin inhibits proliferation of MCF - 7 cells in vitro, which might be related with apoptosis via suppressing the protein expression of PI3K/Akt signaling pathway.
