摘  要:目的 采用免疫组织化学方法对滤泡细胞来源的不同甲状腺组织进行染色并观察碘钠同向转运体(NIS)的表达.方法 收集甲状腺手术切除标本存档蜡块共187例,癌旁甲状腺组织30例为对照,采用链菌素抗生物素蛋白-过氧化物酶(SP)法检测NIS在不同甲状腺组织中的表达.结果 分化型甲状腺癌组织NIS阳性率为12.69%,甲状腺腺瘤NIS阳性率为30.43%,癌旁甲状腺组织NIS阳性率为46.67%,原发性甲状腺机能亢进症组织NIS阳性率为100.00%,甲状腺腺瘤和癌旁甲状腺组织两组比较差异无统计学意义(P>0.05),其余两两比较差异均有统计学意义(P<0.01).结论 NIS蛋白在分化型甲状腺癌的细胞膜上的表达从阴性到强阳性均有分布,NIS蛋白在分化型甲状腺癌的细胞膜上的阳性表达水平可间接预测分化型甲状腺癌细胞的摄碘能力和^131I放射治疗的效果.Objective To observe the expression patterns of sodium/iodide symporter (NIS) in thyroid tissues,discuss the correlation between the expression of NIS in differentiated thyroid cancer and its ability of uptaking iodine,and provide a theoretical basis for ^131I radiation therapy.Methods 187 cases of surgcal resection of patients with thyroid tissue were selected as experimental group,and 30 cases of pericarcinomatous normal thyroid tissues served as control group.The streptavidin-perosidase (SP) method was used to detect the expression of NIS in different thyroid tissues.Results The positive expression rate of NIS in differentiated thyroid cancer,thyroid adenoma,thyroid tissue adjacent to thyroid cancer and primary hyperthyroidism was 12.69%,30.43%,46.67% and 100.00% respectivrely.The staining intensity of NIS in the primary hyperthyroidism was higher than thyroid adenoma and thyroid tissues adjacent to thyroid cancer,and that in the thyroid adenoma and thyroid tissues adjacent to thyroid cancer was higher than in differentiated thyroid cancer.Conclusion The NIS proteins were expressed diversely in cell membrane of differentiated thyroid cancer.This study provides a theoretical basis for individualized ^131I radiation therapy.The positive expression level of NIS protein in cell membrane of differentiated thyroid cancer may indirectly predict the ability of uptaking iodine and the effect of 131I radiotherapy for differentiated thyroid cancer.
