摘  要:目的探讨N—myc下游调节基因1(NDRG1)在前列腺癌(PCa)组织和不同PCa细胞株的表达及其意义。方法应用免疫组织化学染色法检测NDRG1在46例PCa和42例良性前列腺增生(BPH)组织的表达,Westernblot技术检测NDRG1在正常前列腺上皮细胞RWPE1、癌细胞株LNcap和PC-3的表达。结果NDRG1蛋白在BPH组织中阳性表达率95.2%(40/42),在PCa组织中阳性表达率43.5%(20/46),组间差异有统计学意义(P〈0.05)。NDRG1表达与PCa病理分级和临床分期呈负相关,不同组间差异有统计学意义(P〈0.05)。NDRG1蛋白在PC-3和LNcap癌细胞株的表达量均低于其在RWPE1的表达(P〈0.05)。结论NDRG1在PCa组织和不同癌细胞株中的表达均降低,NDRG1的缺失表达可能与前列腺癌的发生、发展有关。Objective To investigate the expression and significance of N - myc downstream - regulated gene 1 ( NDRG1 ) in human prostate cancer (PCa) tissues and different PCa cell lines. Methods The immunohistochemical method was used to detect the expression level of NDRG1 protein in 46 cases of PCa and 42 cases of benign prostatic hyperplasia (BPH) tissues. Western blotting was used to detect the expression volume of NDRG1 protein in normal prostate endothelial cell RWPE1 and PCa cell lines (PC- 3, LNcap). Methods The positive expression rate of NDRG1 protein was 95.2% (40/42) in BPH tissues and 43.5 % (20/46) in PCa tissues ( P 〈 0.05 ). With the increase of clinical stage and path- ological grade of PCa, the positive expression of NDRG1 protein was decreased significantly ( P 〈 0. 05 ). The expression level of NDRG1 protein in PCa cell lines PC -3 and LNcap was lower than that in RWPE1 (P 〈 0.05 ). Conclusion The expression absence of NDRG1 protein may related to the carcinogenesis and progression of PCa.
