摘  要:目的探讨牛磺酸和1,2-二甲基-3-羟基-4-吡啶酮(DFP)联合暴露对铝染毒大鼠学习记忆功能的改善作用。方法将50只健康SPF级雄性Wistar大鼠按体重随机分为5组,分别为阴性对照(1.0 ml生理盐水)组、铝染毒组、牛磺酸干预组、DFP干预组、牛磺酸+DFP干预组,每组10只。采用灌胃方式进行染毒,每天1次,每周连续6 d,持续8周。前4周,铝染毒组、牛磺酸干预组、DFP干预组、牛磺酸+DFP干预组染毒281.40 mg/kg三氯化铝;后4周,铝染毒组给予1.0 ml生理盐水,牛磺酸干预组给予400 mg/kg牛磺酸,DFP干预组给予13.82 mg/kg DFP,牛磺酸+DFP干预组上午给予400 mg/kg牛磺酸,下午给予13.82 mg/kg DFP。利用Morris圆形水迷宫实验测试大鼠学习记忆功能,并测定一氧化氮合酶(NOS)和乙酰胆碱酯酶(ACh E)活力。结果与阴性对照组相比,铝染毒组大鼠Morris水迷宫实验训练期第4天时的总路程和潜伏期均延长,而测试期穿越平台次数减少,大鼠脑组织中NOS活力降低,ACh E活力升高,差异有统计学意义(P<0.05);而各干预组大鼠的总路程、潜伏期和穿越平台次数及大鼠脑组织中NOS和ACh E活力均无明显改变。与铝染毒组相比,DFP干预组、牛磺酸+DFP干预组大鼠的总路程和潜伏期均缩短,穿越平台次数增加;各干预组大鼠脑组织中NOS的活力升高,ACh E的活力降低,差异有统计学意义(P<0.05)。与牛磺酸、DFP干预组相比,牛磺酸+DFP干预组大鼠的总路程和潜伏期均缩短,穿越平台次数增加;大鼠脑组织中NOS的活力升高,ACh E的活力降低,差异均有统计学意义(P<0.05)。结论牛磺酸与DFP通过保护脑组织神经系统而改善铝染毒大鼠学习记忆功能,两者联合暴露较单一暴露的效果更为显著。Objective To investigate the protective effects of taurine and DFP in combination on learning and memory of rats exposed to aluminum. Methods Fifty male Wistar rats were randomly divided into five groups with ten in each group,control group(1.0 ml normal saline),aluminum group,taurine group,DFP group,combination group(taurine +DFP),respectively. The tested chemicals were given by gavage six days per week with one time each day for eight weeks. The aluminum group,taurine group,DFP group and the combination group were administered with aluminum chloride(281.40 mg/kg body weight) in the first four weeks. In the last four weeks,the aluminum group and control group were given 1.0 ml/d saline,respectively. The taurine group was given 400 mg/(kg·d) taurine and the DFP group was given 13.82 mg/(kg·d) DFP. The combination group was given 400 mg/(kg·d) taurine in the morning,and 13.82 mg/(kg·d) DFP in the afternoon. The Morris circle water labyrinth was used to test learning and memory of rats and the contents of NOS and ACh E were tested. Results Compared with the control group, the total distance and latency of rats exposed to aluminum in the fourth day in Morris water maze test were both longer,with less number of times through the platform in the testing stage; The levels of NOS and ACh E in brains of rats exposed to aluminum were lower and higher, respectively, both with statistically difference(P〈0.05). However, it was not significant in the change of the total distance, latency, the number of times through the platform and the levels of NOS and ACh E in brains of rats in the intervention groups. Compared with the aluminum group,the total distances and latency of rats in the DFP group and the combination group were both shorter, with more number of times through the platform; The levels of NOS and ACh E in brains of rats exposed to aluminum in each intervention group were lower and higher respectively(P〈0.05).
