异嗪皮啶对肝癌患者MMP-7表达水平和癌细胞侵袭能力的影响

摘　　要：目的探讨异嗪皮啶对肝癌患者基质金属蛋白酶7(MMP-7)表达水平和癌细胞侵袭能力的影响。方法将126例肝癌患者根据治疗方法不同分为常规治疗组和异嗪皮啶组,各63例。两组患者均给予化疗、手术等治疗,异嗪皮啶组给予异嗪皮啶治疗,依据使用剂量分为高亚组(0.35 mg/kg)、中亚组(0.25 mg/kg)和低亚组(0.15 mg/kg)。随访3年,统计分析并采用免疫组化法检测所有患者MMP-7表达水平、流式细胞法监测癌细胞周期动力学(G0/G1、S、G2M期)和应用生存曲线分析生存预后水平。结果治疗后,异嗪皮啶组患者MMP-7表达阳性率较治疗前明显下降(P<0.01),且异嗪皮啶组表达阳性率低于常规治疗组(P<0.01);异嗪皮啶组患者G0/G1期和S期癌细胞周期动力学均低于常规治疗组(P<0.05),G2M期高于常规治疗组(P<0.05)。治疗后,高亚组和中亚组均较治疗前明显下降(P<0.01);G0/G1期和S期癌细胞周期动力学比较,高亚组<中亚组<低亚组(P<0.05);G2M期癌细胞周期动力学比较,高亚组>中亚组>低亚组(P<0.05)。生存曲线分析结果显示,异嗪皮啶组3年内总生存率高于常规治疗组(P<0.05)。结论异嗪皮啶可有效抑制肝癌患者MMP-7和癌细胞生长,降低癌细胞侵袭能力和提高生存预后,值得临床推广应用。Objective To discuss the effect of Isofraxidinin on expression of matrix metalloproteinases 7（MMP-7）levels and cancer cell invasion ability of patients with liver cancer.Methods One hundred and twenty six patients with liver cancer were divided into routine treatment group and Isofraxidinin group according to different treatments,63 in either group.Patients in both group were given chemotherapy and operative treatment,and patients in Isofraxidinin group were given Isofraxidinin treatment and on the basis of dosage were divided into high subgroups（0.35 mg / kg）,middle subgroup（0.25 mg / kg）and low subgroups（0.15 mg / kg）.After 3 years＇ follow-up,MMP-7 expression levels of all patients were detected and analysed.Flow cytometry method was used to monitor cancer cell growth situation（G0 / G1,S,G2 M phase）prognosis and survival curve were used to analyze the survival level.Results After treatment,Positive rate of MMP-7 expression of patients in Isofraxidinin group greatly decreased（P 〈0.01）and positive rate of cancer cell growth was significantly lower than that of the conventional treatment group（P〈 0.01）.Cancer cell growth in G0 / G1 and S of patients in Isofraxidinin group was lower than that of conventional treatment group,but the growth in G2 M was higher（P〈 0.05）.After treatment,positive rate of cancer cell growth in middle subgroup and high subgroup both decreased（P 0.01）.Survival curve analysis showed that,patients＇ three years overall survival rate in Isofraxidinin group was obviously higher than that of conventional treatment group（P〈 0.05）.Conclusion Isofraxidinin can effectively inhibit MMP-7 and cancer cell growth of patients with liver cancer,reduce the cancer cell invasion ability and improve prognosis,so it is worth for further clinical promotion.
