黑蒜提取液对胰腺癌Panc-1细胞生长与抗氧化分子机制的实验研究

摘　　要：目的探讨黑蒜提取液(aged black garlic extract,ABGE)对Panc-1细胞生长与抗氧化能力的影响。方法将不同浓度的ABGE(0.2、1.0、1.8 mg/ml)作用于Panc-1细胞,采用CCK-8法分别于24 h、48 h、72 h测定Panc-1细胞株的生长能力;RT-PCR检测ABGE干预后Panc-1细胞VEGFm RNA以及MMP-9m RNA表达变化;ELISA法检测Panc-1细胞上清液中相关蛋白VEGF、SOD的表达情况。结果 CCK-8结果显示,黑蒜提取液对Panc-1细胞株的生长活性具有明显的抑制作用,且具有时间剂量依赖关系;RT-PCR结果显示,黑蒜提取液能明显地上调VEGFm RNA以及下调MMP-9m RNA的表达;ELISA法结果显示,随黑蒜提取液浓度的增加,Panc-1细胞上清液中VEGF蛋白的表达水平下降、SOD蛋白表达增加。结论黑蒜提取液能显著抑制Panc-1细胞的生长能力,其抗氧化能力可能是与下调VEGF蛋白表达、MMP-9以及上调SOD表达有关。Objective To explore molecular mechanism of aged black garlic extract（ABGE） on pancreatic cancer-1 cell growth and antioxidant ability. Methods The different concentrations of ABGE（0.2,1.0,1.8 mg / ml）acted as Panc-1 cell,the growth activity of Panc-1 was detected by CCK-8 at 24,48 and 72 h;the VEGF and SOD in supernatant of Panc-1 was detected by ELISA method;and the VEGFm RNA and MMP-9m RNA protein expression of Panc-1 was detected by Real-time-PCR. Results CCK-8 test showed that ABGE had the inhibitory effect on Panc-1 and in a time-dose-dependent manner;ELISA method showed that down-regulating VEGF and up-regulating SOD protein expression;Real-time-PCR test showed that ABGE could obviously downregulate VEGF and MMP-9 protein expression. Conclusion Aged black garlic extract can significantly inhibit pancreatic cancer-1 cell growth and tranfer,which may be the cause ABGE could interfere TGF-β1 / Smad4 signaling pathway.
