摘 要:目的探讨丁苯酞联合依达拉奉对局灶性脑梗死大鼠皮质血脑屏障及咬合蛋白表达的影响。方法选取成年SD雄性大鼠60只,随机分为对照组、模型组、丁苯酞组、丁苯酞联合依达拉奉组,每组15只。各组于模型制作成功后72h采用伊文思兰渗出法检测大鼠血脑屏障通透性变化,免疫组织化学和Western blot检测损伤区周围皮质咬合蛋白表达变化。结果与对照组比较,模型组、丁苯酞组、丁苯酞联合依达拉奉组梗死区大脑皮层血脑屏障通透性均增加,皮质咬合蛋白表达均减少。与模型组相比,丁苯酞组和丁苯酞联合依达拉奉组皮质脑组织伊文思蓝渗出量减少,皮质咬合蛋白表达增加。与丁苯酞组比较,丁苯酞联合依达拉奉组伊文思蓝渗出量减少,咬合蛋白表达量增加。结论丁苯酞软胶囊可保护血脑屏障,增加大脑皮质咬合蛋白的表达。与依达拉奉联合应用保护血脑屏障、增加咬合蛋白表达作用更明显。Objective To explore the effects of butylphthalide combined with edaravone on blood-brain barrier permeability and occludin expressions in cortex of focal cerebral infarction in rats.Methods Sixty sprague-dawley male rats were randomly divided into four groups:control group,model group,butylphthalide group and butylphthalide combined with edaravone group,with 15 rats in each group.The model was established based on the principle of photochemical initiation of thrombosis.At 72 hafter injury,evans blue extravsation method was used to detect the blood-brain barrier permeability.Immunohistochemistry and western blot were used to examine the cortex occludin expression around the infarction area.Results Compared with the control group,the blood-brain barrier permeabilities in the model group,butylphthalide group and the butylphthalide combined with edaravone group were increased significantly.And the occludin expressions aroud the infarction were upregulated significantly.Compared with the model group:evans blue extravsations in cortex in the butylphthalide group and in the butylphthalide combined with edaravone group were decreased,and the occludin expressions aroud the infarction area were increased.Compared with the the butylphthalide group:evans blue extravsation in cortex in the butylphthalide combined with edaravone group was decreased,and the occludin expression aroud the infarction was increased.Conclusion Butylphthalide could protect the bolld-brain barrier,and increase the expression of occludin.The effect was more obvious by using the combination of butylphthalide and edaravone.
