摘  要:目的探讨COPD大鼠模型中气道弹性及炎症因子的改变。方法将40只Wistar大鼠随机分为5组,每组再分两组(COPD模型组及正常对照组),采用烟熏加气管内注入脂多糖的方法制造COPD大鼠模型。用HE三染色及马松(Masson)三色染色评价大鼠气道壁病理变化和胶原纤维的变化;采用ELISA法检测血清中基质金属蛋白酶9(matrix metalloproteinase9,MMP9)、金属蛋白酶抑制物1(tissue inhibitor of metalloproteinasel,TIMPl)及热休克蛋白(heatshock proteins27,HSP27)的表达水平。结果COPD模型组血清MMP9、TIMP1及HSP27的表达水平高于正常对照组的表达水平;COPD模型组MMP9的表达总体呈上升趋势,差异有统计学意义(P〈0.01);COPD模型组TIMP1的表达总体呈上升趋势,G3与G5组(P〈0.01)及G4与G5组(P〈0.01)差异有统计学意义;COPD模型组MMP9/TIMP1的比值随MMP9和TIMP1改变而变化,差异有统计学意义(P〈0.01);COPD模型组HSP27的表达呈轻微上升趋势,G4与G5组之间差异有统计学意义(P〈0.05)。结论MMP9/TIMP1的平衡及胶原纤维、HSP27的改变可以作为评价COPD进展的重要指标。Objectives To investigative the variation of airway flexibility and inflammation factors in the analogue of rats with COPD.Methods 40 Wistar rats were derided into five groups randomly, eath group was derided into two groups: COPD model group and normal control group.The rats' COPD model were made by smoking and lipopolysaccharide injected in the trachea.To appreciation the change of pathology of rats' airway wall and collagen by HE and Masson.To detect the level of MMPg, TIMP1 and HSP27 by ELISA.Results The level of MMPg, TIMP1 and HSP27 in COPD model groups appeared higher than those of nomal control group.The level of MMP9 in COPD model groups presented a trend of rising on the whole,difference was statistically significant (P〈0.01) ;the level of TIMP1 in COPD model groups also showed rising trend as a whole,difference between G3 and G5 (P 〈 0.01 ) and difference between G4 and G5 (P 〈 0.01) was statistically significant;the ratios of MMPg/TIMP 1 changed as the level of MMP9 and TIMP1 change,difference was statistically significant (P〈0.01) .The level of HSP27 in COPD model groups had a slightly rising tend,difference between G4 and G5 was statistically significant(P〈0.05 ) .Conclusions The balance of MMPg/TIMP1 and the change of collagen and HSP27 cold as a new approach to COPD control.
