摘 要:为了提高炼油废水生物处理装置的反硝化性能,从武汉石化污水处理厂的活性污泥中筛选出一株好氧反硝化菌AD10,经生理生化及16S r DNA序列分析,将其鉴定为假单胞菌属。通过单因素实验研究了该菌的最适培养条件:以丁二酸钠为碳源,C/N=14,温度30℃,初始p H=6.0,摇床转速200r/min。在此条件下,ρ(NO_3^--N)=556.81mg/L时72h的去除率为97.2%,且耐受的硝酸盐氮质量浓度达到654.00mg/L。菌株AD10培养11.5 h,对ρ(NO_3^--N)=140.31mg/L和ρ(TN)=141.62mg/L的去除率分别为95.8%和93.9%,其反硝化速率高于大部分已发现的好氧反硝化菌,检测到的ρ(NO_2~–-N)≤2.30mg/L,未出现明显的积累,菌株AD10能够进行完全反硝化。In order to improve the denitrification efficiency of the bioreactor of refinery wastewater treatment plant,an aerobic denitrifier AD10 was isolated from the activated sludge of a petrochemical wastewater treatment plant in wuhan city. It was identified as a member of the genus Pseudomonas sp. according to its morphyological and physiological properties and 16 S r DNA gene sequence analysis. Through the single factor experiment investigations,the optimum denitrification conditions were: sodium succinate as carbon source,C/N 14,temperature 30℃,initial p H 6.0,rotation speed 200 r/min. Under these conditions,the nitrate remove efficiency of AD10 was 97.2% when it was cultivated in 556.81 mg/L nitrate for 72 h,and it could tolerate at least 654 mg/L of nitrate nitrogen. Culturing after 11.5h,AD10 show the NO_3~–-N and TN removal rate of 95.8% and 93.9%,when the concentration of NO_3^--N and TN were 140.31mg/L and 141.62mg/L,respectively. AD10's denitrification rate is higher than most of the aerobic denitrifying bacteria have been found. During this process,only a very small amount of NO_2~–-N(≤2.30mg/L) was detected,indicating a complete denitrification.
