奥美沙坦对左心室肥厚大鼠肾脏的保护作用

摘　　要：目的研究奥美沙坦对心肌肥厚大鼠肾脏Ang II、TGFβ-1水平的作用及与肾小管间质纤维化的关系。方法60只Wistar大鼠随机分三组,模型组和干预组0.02mg/(kg·d)异丙肾上腺素,bid,H,连用10天;干预组奥美沙坦10 mg/(kg·d)加蒸馏水1ml,余两组1ml蒸馏水,qd灌胃;笼养8周后B超测定大鼠肾脏大小;检测肾动脉阻力指数及Ang II、TGFβ-1水平。结果对照组肾动脉阻力指数均明显低于其他两组;干预组明显低于模型组;Ang II、TGFβ-1及间质胶原的含量差异明显。结论奥美沙坦可以抑制后超负荷压力增加引起的肾间质纤维化。Objective To observe the effects of olmesartan on Angiotensin II（ Ang II）,transforming growthfactor Bl,（ TGF- β1） in cardiac hypertrophy rat,and the relation of the degree of tubulointerstitial fibrosis. Methods Totally 60 Wistar rats were randomly divided into three groups： control group and isoprenaline group. When had been established as the model,the rats of isoprenaline group were randomly divided into model group and intervention group. Based on standard feeding method,control group the rat subcutaneous injection of 1ml saline bid. The model group and intervention group in the rat subcutaneous injection of isoproterenol 0. 02 mg /（ kg·d） bid. 1. 0 ml double distilled water was given through intragastric administration to each rat every day by group. And 10 mg /（ kg·d） dose of olmesartan was contained in the double distilled water which was administrated to the rats of intervention group,no drug was given in the other two groups. The rats were killed by groups after eight weeks intragastric administration,and then measured Ang II,TGFβ- 1 concentration levels of kidney tissue,comparison of measured values of kidney size in rats,renal artery resistance index. Results The renal artery resistance index infarction group and intervention group were significantly higher than that in control group,and significantly lower in intervention group vs model group; the concentration levels of Ang II and TGFβ- 1 in kidney tissue had positive correlation with the degree of renal tubulointerstitial fibrosis. Conclusion Olmesartan inhibits induced renal interstitial fibrosis
