摘  要:为了筛选槐猪和杜洛克猪背最长肌中的差异表达基因,对槐猪和杜洛克猪背最长肌进行转录组测序,并进行生物信息学分析.结果表明,槐猪和杜洛克猪分别获得36 177 844和34 496 450条可用读段,且与猪参考基因组的比对率分别为78.71%和80.09%.槐猪和杜洛克猪间共有953个差异表达基因,以杜洛克猪为对照组,槐猪与之相比,有589个上调基因和364个下调基因.槐猪和杜洛克猪的差异表达基因共分类到48个基因本体(GO)条目中,其中,细胞组分13个、分子功能12个和生物学过程23个.显著富集的KEGG通路共计6个,其中,PPAR信号通路是与调节脂类代谢显著相关的通路.In order to investigate the function of differentially expressed genes and gene product on metabolism,digital gene expression profiles in longissimus dorsi muscle of Huai pig( fat) and Duroc pig( lean) were built using RNA-seq method,and differentially expressed genes were screened and annotated. Gene ontology( GO) classification analysis and significant enrichment analysis of Kyoto encyclopedia of genes and genomes( KEGG) pathway were conducted. Results showed that 36 177 844 and 34 496 450 reads were obtained from Huai pig and Duroc pig,respectively. A total of 953 differentially expressed genes were detected between two breeds and were classified into 48 GO terms. Among the terms,13 terms were in cellular component ontology,12 terms in molecular function ontology and 23 terms in biological process ontology. Compared to Duroc pig,Huai pig had 589 up-regulated genes and 364down-regulated genes. All differentially expressed genes were in 6 enriched KEGG pathways,with PPAR signaling pathway relating to lipid metabolism.
