摘  要:目的:探讨脾气虚模型大鼠焦虑样行为与海马血管活性肠肽的关系。方法:16只SD大鼠随机分为对照组和模型组,每组各8只;旷场实验评价模型组大鼠的焦虑程度;ELISA法检测两组大鼠的海马组织血管活性肠肽(VIP)水平;免疫组织化学法观察两组大鼠的海马组织血管活性肠肽1,2受体(VPAC1,2R)的表达,并进行组间比较。结果:与对照组比较,模型组大鼠的中心活动时间明显延长,周围活动时间显著减少;海马VIP水平无明显变化,但VPAC1,2R表达显著上升。结论:脾气虚可降低大鼠的焦虑程度,其机制可能与大鼠海马组织的VPAC1,2R表达上调有关。Objective:To study anxiety-like behaviors of spleen qi deficiency rat and their relations with hippocampal vasoac-tive intestinal peptide ( VIP) and its receptors ( VPACR) . Methods:16 SD rats were randomly divided in control group and spleen qi deficiency group ( model group) , 8 rats each group. The open field test was used to explore the anxious behaviors of the rats;ELISA was employed to measure concentrations of hippocampal VIP;and immohistochemistry was used to observe expressions of VPAC1/2R. The results of the two groups were compared. Results:Compared with those in the control group, the model rats showed longer central duration and shorter peripheral duration, exhibited unchanged VIP level in hippocampus, and greater expressions of hippocampal VPAC1/2R. Conclusions:Spleen qi deficiency can reduce anxiety-like behaviors and its underlying mechanism is relative to the up-regulation of hippocampal VPAC1/2R expressions.
