摘 要:目的研究肝糖原在小鼠酒精性肝损伤过程中的动态变化及意义。方法 60只雄性小鼠随机分为2组:正常组(n=10),模型组(n=50)。正常组小鼠直接处死取肝;模型组小鼠每天给予56度红星二锅头(15 m L·kg^(-1)),连续灌胃5周,建立酒精性肝损伤模型。于建模第1,2,3,4,5周,各处死10只,取肝组织。用糖原过碘酸雪夫氏(PAS)染色,检测各组肝组织切片中肝糖原的表达情况即平均光密度值(MOD)。结果与正常组的MOD值(0.22±0.04)相比,模型组第1周(0.19±0.06 vs 0.22±0.04,P<0.01),第2周(0.17±0.05 vs0.22±0.04,P<0.01),第3周(0.14±0.02 vs 0.22±0.04,P<0.01),可见肝糖原含量依次下降,至第3周(0.14±0.02)肝糖原含量最低;而模型组第4周(0.17±0.07 vs 0.14±0.02,P<0.01),第5周(0.19±0.03 vs 0.17±0.07,P<0.05)肝糖原含量呈上升趋势。结论小鼠酒精性肝损伤过程中,肝糖原的变化规律为先减少后增加。Objective To investigate the expression changes and significance of hepatic glycogen during liver injury of mice induced by alcohol.Methods Sixty clean and healthy male mice were randomly divided into two groups: normal group( n = 10) and model group( n = 50). All animals in normal group were executed to obtain the liver tissue. The model mice were administered with alcohol of 56 degrees( 15 mL·kg^-1) for five weeks to induceliver injury,ten of which were given to execute after feeding 1,2,3,4,5 weeks,respectively. The expression levels of hepatic glycogen in all groups were detected by periodic acid- schiff( PAS) staining( MOD). Results Compared to the normal group [MOD,( 0. 22 ±0. 04) ],the levels of hepatic glycogen were decreased gradually in the model group at 1- week(0. 19 ± 0. 06),2- week( 0. 17 ± 0. 05),at 3- week( 0. 14 ± 0. 02; all P〈0. 01) was the lowest. Compared to the mice in model group at 3- week( 0. 14 ± 0. 02),the hepatic glycogen contents were increased significantly at 4- week( 0. 17 ± 0. 07; P〈0. 01) and 5- week( 0. 19 ± 0. 03; P〈0. 05). Conclusion In the process of alcohol- induced liver injury in mice,the levels of hepatic glycogen were decreased gradually at the early stage,and then increased in the late.
