摘  要:目的检测大鼠脑缺血再灌注后海马神经细胞损伤及自噬变化,并比较海马CA_1及CA_3区的自噬情况。方法用线栓法制作大鼠脑局部缺血再灌注模型,实验动物随机分为:对照组(Control组)、假手术组(Sham组)和大脑中动脉栓塞再灌注组(MCAO组)。HE染色检测大鼠海马神经细胞损伤。透射电镜下观察海马神经元内自噬小体。免疫荧光法检测自噬相关蛋白LC3、Beclin-1的表达水平。结果与Control组及Sham组比较,大鼠脑缺血再灌注后,海马神经细胞损伤严重;海马神经元内有自噬体形成;自噬标记物LC3、Beclin-1蛋白表达水平显著增加,且CA_1区的表达量明显高于CA_3区(差异均具有统计学意义P<0.05)。结论脑缺血再灌注可激活海马神经细胞内的自噬,并进一步引起细胞损伤;模型动物海马CA_1区的自噬水平显著高于CA_3区,暗示CA_1区对脑缺血损伤具有较高的敏感度。Objective To investigate the damage of hippocampal neural cells,the changes of autophagy and the difference of autophagy between the CA1 and CA_3 region of hippocampus after cerebral ischemia-reperfusion injury in rats.Methods The focal cerebral ischemia-reperfusion model was induced by an intraluminal filament embolism. The SD rats were randomly divided into 3 groups: the control group( Control group),the sham-operated group( Sham group) and the middle cerebral artery occlusion- reperfusion group( MCAO group). HE staining was employed to detect the damage of hippocampal neurons in rats. The formation of autophagosomes in hippocampal neurons was observed by using transmission electron microscope. The expression levels of autophagy related protein Beclin-1 and LC3 in hippocampus CA_1 and CA_3 region was detected by immunofluorescence assay. Results The hippocampal neurons in ischemia brains were damaged seriously and the formation of autophagosomes were detected by transmission electron microscope. Compared with Control group and Sham group,the results by immunofluorescence showed that the expression of LC3 and Beclin-1 protein in hippocampus neurons were significantly increased,and the expression in CA_1 region was significantly higher than that in CA_3 region( P〈0. 05). Conclusion Cerebral ischemia-reperfusion can activate autophagy in hippocampal neurons,and further cause cell damage. The level of autophagy of CA_1 region of the model animals was significantly higher than that of CA_3 region,suggesting that CA_1 region had a high sensitivity to cerebral ischemia injury.
