微小RNA-146a rs2910164及微小RNA-499 rs3746444与缺血性脑卒中的关系

摘　　要：目的探讨微小RNA（miR）-146a rs2910164和miR-499 rs3746444单核苷酸多态性与缺血性脑卒中的关系。方法采用基因直接测序技术及Taqman-PCR基因分型技术检测378例缺血性脑卒中患者（病例组）及378名健康人（对照组）miR-146a rs2910164和miR-499 rs3746444位点的等位基因和基因型的频率分布,采用条件Logistic回归分析其与缺血性脑卒中的关系。结果病例组的miR-146a rs2910164位点等位基因G频率分布高于对照组（P=0.003,OR=1.412,95%CI=1.121~1.716）,基因型GG及CG＋GG的频率分布也高于对照组（GG：P=0.002,OR=2.080,95%CI=1.330~3.250;CG＋GG：P=0.011,OR=1.488,95%CI=1.110~2.012）。2组miR-499rs3746444位点的等位基因与基因型频率分布比较差异均无统计学意义（P均〉0.05）。结论 miR-146a rs2910164单核苷酸多态性可能与缺血性脑卒中相关,miR-499 rs3746444则可能与缺血性脑卒中无关。Objective To investigate the association of miR-146 a rs2910164 and miR-499rs3746444 single nucleotide polymorphisms and ischemic stroke. Methods Taqman-PCR and DNA sequencing assays were performed to investigate the genotype and allele gene of miR-146 a rs2910164 and miR-499rs3746444 loci in 378 patients with ischemic stroke（ ischemic stroke group） and 378 healthy individuals（ control group）. The association of miR-146 a rs2910164 and miR-499 rs3746444 single nucleotide polymorphisms with the risk of ischemic stroke was assessed by logistic regression analysis. Results In the ischemic stroke group,the frequency of allele G（ P = 0. 003,OR = 1. 412,95% CI = 1. 121-1. 716）,genotype GG（ P =0. 002,OR = 2. 080,95% CI = 1. 330-3. 250）,CG ＋ GG（ P = 0. 011,OR = 1. 488,95% CI = 1. 110-2. 012）of miR-146 a rs2910164 was significantly higher compared with those in the control group. However,the frequency of allele gene and genotype of miR-499 rs3746444 did not significantly differ between the ischemic stroke and control groups（ both P 〉0. 05）. Conclusions The single nucleotide polymorphism of miR-146 a rs2910164 rather than miR-499 rs3746444 is probably correlated with the risk of ischemic stroke.
