摘 要:目的揭示松多酚(pine polyphenols,PPs)及其炭黑曲霉转化产物(Aspergillus carbonarius,ACPP)对γ-辐射诱导小鼠免疫损伤的防护机制。方法以ACPP和PPs为研究对象,比较对ABTS,DPPH,超氧阴离子和羟基自由基的清除活性。采用60Coγ-射线诱导的小鼠模型,MTT和生化分析法对外周血象、吞噬指数、脾B淋巴细胞转化增殖及脾组织的生化指标进行分析,蛋白质印迹法分析脾Bax,Bcl-2和Caspase-3表达水平。结果 ACPP显示更强的抗氧化活性。ACPP和PPs主要通过提高体内抗氧化物质含量来促进酶(SOD,CAT,GSH-Px)和非酶抗氧化剂(GSH)、脾B淋巴细胞转化增殖和单核细胞的吞噬作用,降低脂质过氧化水平;降低Bax和Caspase-3表达,增加Bcl-2表达起到辐射防护作用。ACPP防护效果明显优于PPs。结论 ACPP具有抗氧化活性和辐射防护作用,是一种可靠的抗辐射基料。Objective To reveal the protective mechanism of the immune injury in mice induced by γ-radiation of pine polyphenols( PPs) and its transformation by Aspergillus carbonarius( ACPP). Methods ACPP and PPs served as the research subjects. The scavenging activities of ABTS,DPPH,superoxide anion and hydroxyl free radicals were assayed and compared. The mice model induced by60 Co γ-radiation was adopted and the spleen B lymphocyte transformation proliferation,peripheral blood and biochemical status was determined by MTT and biochemical methods. The expression levels of Bax,Bcl-2 and Caspase-3 were determined by Western blotting. Results ACPP demonstrated a stronger antioxidant activity. The radioprotective effect of ACPP and PPs was realized by improving the content of antioxidants,promoting enzymatic( SOD,CAT and GSHPx) and non-enzymatic( GSH) antioxidants,spleen B lymphocyte transformation proliferation and phagocytic index,reducing the lipid peroxidation and the expression levels of Bax and Caspase-3,and increasing the Bcl-2 in the spleen tissue. The radioprotective effect of ACPP was better than PPs. Conclusion ACPP is of antioxidant activity and radioprotective effect,and so it is a reliable basic material in resistance to ionizing radiations.
