摘 要:目的:观察白藜芦醇对A549肺腺癌细胞生长的影响并对促凋亡机制进行初步研究。方法:使用MTT方法观察不同浓度与作用时间的白藜芦醇对A549细胞增殖的影响,采取流式细胞技术观察白藜芦醇对A549细胞促凋亡情况与细胞周期的影响,选用Western-blot技术观察不同浓度白藜芦醇作用下A549细胞凋亡相关蛋白p53、Bcl-2家族及Caspase的变化。结果:白藜芦醇对A549细胞增殖的抑制呈浓度/时间双重依赖,作用24 h IC50值是29.3μM,这种抑制作用有凋亡机制的参与。A549细胞发生了S期阻滞,凋亡相关蛋白中p53浓度增加,Bcl-2家族蛋白中促凋亡蛋白/抗凋亡蛋白比值上调,Caspase-3、-9发生剪切。结论:白藜芦醇能通过促凋亡作用抑制A549肺腺癌细胞的增殖,诱导出现S期细胞阻滞,与细胞凋亡线粒体途径相关的p53、Bcl-2家族蛋白、Caspase-3、-9等多种凋亡相关因子也参与促凋亡作用中,白藜芦醇对A549的促凋亡效应可能是一个多靶点多层次的作用机制。Objective: To observe the effects of resveratrol on the growth of A549 lung adenocarcinoma cells and preliminarily invest the molecular mechanism. Methods: The anti- proliferation effect of resveratrol on A549 cells was detected by using MTT assay. Flow cytometry was used to observe the influence of resveratrol on the apoptosis and the cell cycle of A549 cells. Western- blot technique was employed to study the effects of different concentrations of resveratrol to the changes of p53 and its downstream targets,such as Bcl- 2 family and caspases,in A549 cells. Results: Resveratrol could inhibit the proliferation of A549 cells in a dose- and time- dependent manner. When A549 cells were treated with resveratrol for 24 h,its estimated IC50 was 29. 3 μM. A549 cells were arrested in the S phase. Resveratrol also upregulated the activity of p53 and modulated the balance of pro-/anti- apoptotic molecules in Bcl- 2 family and promoted the cleavage of Caspase- 3 and Caspase- 9. Conclusion: Our findings suggested that resveratrol could induce the apoptosis and inhibit the proliferation of A549 cells. After treatment of resveratrol,A549 cells were arrested in the S phase. Meanwhile,p53,Bcl- 2 family and Caspase- 3 and Caspase- 9,which were related with mitochondria- mediated apoptotic pathway,were also involved in. The apoptosis promoting effect of resveratrol on A549 may be through the mechanism of multi- levels and multi- targets.
