摘  要:目的:研究小干扰RNA(small interfering RNA,siRNA)沉默分化抑制因子(inhibitor of differentiation,Id)1基因后骨肉瘤细胞增殖、迁移、侵袭生物学行为的变化。方法:培养骨肉瘤HEK293细胞株,分别转染Id1-siRNA、NC-siRNA,转染24、48h后测定细胞的增殖、迁移和侵袭,转染48h后测定细胞中促增殖基因、增殖抑制基因、迁移和侵袭相关基因的表达量。结果:转染后24、48h,Id1-siRNA组细胞的OD值、侵袭细胞数目均显著低于NC-siRNA组(P<0.05),划痕面积显著大于NC-siRNA组(P<0.05);转染后48h,Id1-siRNA组细胞的hnRNP A2、hnRNP B1、S100A6、Runt相关转录因子2(RUNX2)、Aurora-A、Grb2、Gab2、Rap2a表达量显著低于NC-siRNA组(P<0.05),p53、TAp73、IGFBP5显著高于NC-siRNA组(P<0.05)。结论:siRNA沉默Id1基因能够抑制骨肉瘤细胞的增殖、迁移、侵袭。Objective:To study the changes of osteosarcoma cell biological behaviors proliferation,migration and invasion after inhibitor of differentiation 1(Id1)gene silencing by small interfering RNA(siRNA).Methods:Osteosarcoma HEK293 cell lines were cultured and transfected with Id1-siRNA and NC-siRNA respectively,cell proliferation,migration and invasion were detected after 24 hand 48hof transfection,and the expression levels of proliferation-promoting genes,proliferation-inhibiting genes as well as migration and invasion-related genes in cells were detected after 48 hof transfection.Results:After 24 hand 48hof transfection,cell OD value and the number of invasive cells of Id1-siRNA group were significantly lower than those of NC-siRNA group(P〈0.05),and scratch area was significantly larger than that of NC-siRNA group(P〈0.05);after 48 hof transfection,hnRNP A2,hnRNP B1,S100A6,RUNX2,Aurora-A,Grb2,Gab2 and Rap2aexpression levels of Id1-siRNA group were significantly lower than those of NC-siRNA group(P〈0.05),and p53,TAp73 and IGFBP5were significantly higher than those of NC-siRNA group(P〈0.05).Conclusions:Id1gene silencing by siRNA can inhibit osteosarcoma cell proliferation,migration and invasion.
