摘  要:目的 :检测丙酮酸羧化酶(pyruvate carboxylase,PC)在上皮性卵巢癌中的表达水平及其临床病理学意义,并进一步探讨沉默PC基因的表达对卵巢癌SKOV3细胞增殖及迁移的影响。方法 :分别采用蛋白质印迹法和免疫组织化学方法检测PC在上皮性卵巢癌及其相应的癌旁组织中的表达水平;统计分析PC表达与患者年龄、有无腹腔积液、临床TNM分期及病理分级等临床病理特征之间的关系。通过构建特异性针对PC基因的PC-shRNA沉默SKOV3细胞中PC的表达水平,并采用MTT法、二维克隆形成实验、软琼脂克隆形成实验及细胞迁移实验,观察沉默PC表达对SKOV3增殖及迁移能力的影响。结果 :PC在卵巢癌组织中的表达水平明显高于其对应的癌旁组织(t=20.4,P<0.01);PC蛋白高表达与患者发生腹腔积液和临床分期有关(P值均<0.05)。沉默PC的表达可以显著抑制SKOV3细胞的增殖和迁移能力(P值均<0.01)。结论 :PC在上皮性卵巢癌组织中高表达,PC与卵巢癌的发生和发展密切相关,可作为卵巢癌临床治疗的潜在靶点。Objective: To study (PC) in epithelial ov significance, and to proliferation and mig the expre arian canc explore th ration abili ssion level of pyru er (EOC) and its cl e effect of PC gene ty of ovarian cancer vate carboxylase inicopathological -silencing on cell SKOV3 cells.Methods: The expression levels of PC in EOC tissues and the corresponding para-cancerous tissues were detected by Western blotting and immunohistochemistry. The correlations of PC expression with the clinicopathological features (including age, ascites, clinical stage and pathological grade) of EOC patients were studied using statistical analysis. The expression of PCgene in SKOV3 cells was silenced using PC-shRNA specifically targeting PC gene. Then the effects of PC gene-silencing on cell proliferation and migration ability were detected by MTT assay, two-dimensional clone formation assay, soft agar colony formation assay and wound healing assay, respectively. Results: The expression level of PC in EOC tissues was significantly higher than that in the corresponding para-cancerous tissues (t = 20.4, P 〈 0.01). The high expression of PC was significantly correlated with the occurrence of ascites and clinical stage (both P 〈 0.05). Silencing of PC gene could significantly inhibit cell proliferation and migration of SKOV3 cells (both P 〈 0.01 ). Conclusion: The expression of PC is high in EOC tissues, and PC is closely related to the occurrence and development of EOC. PC may be used as a potential target for clinical therapy of EOC.
