摘 要:目的:研究Poly I:C(Polyinosinic-polycytidylic acid)对急性期溃疡性结肠炎模型小鼠免疫机制调节作用。方法:30只小鼠随机分为正常组(control)、模型组(model)、Poly I:C组(Poly I:C),建立急性溃疡性结肠炎动物模型;观察小鼠疾病活动指数(DAI)、结肠病理学变化、流式细胞术检测T淋巴细胞亚群变化、免疫比浊法检测血清免疫球蛋白变化、免疫组织化学染色测定结肠组织ICAM-1变化;结果:经Poly I:C治疗后模型小鼠的疾病活动指数及病理组织评分明显下降;与model组比较,Poly I:C组免疫细胞反应明显减轻弱,结肠组织ICAM-1蛋白表达明显降低。结论:可见Poly I:C对急性溃疡性结肠炎模型小鼠有免疫调节作用。Objective:To investigate the effects of Poly IC on the immunologic mechanism in DSS-induced acute colitis.Method:Thirty of mice were randomly grouped as follows: control group, model group, Poly IC group, acute colitis were established, disease activity index (DAI) and colon pathological changes were observed,the changes of T lymphocyte subsets and immunoglobulin were detected by Flow cytometry and Immune turbidimetry, the expressions of ICAM-1 proteins were detected by immunohistoehemistry,Results:The DAI and colon pathological were changed in the Poly hC group compared with that of the model group; Immunologic cells were decreased, the proteins expressions of ICAM-1 significantly decreased in the Poly I:C group compared with that of the model group by immunohistochemical staining.Conclusions:The administration of PolyI:C may be adjusted the immunologic mechanism in the acute experimental colitis.
