摘  要:目的探讨生后早期双眼形觉剥夺对大鼠视觉传导通路电生理学特性的影响。方法 50只8 d龄健康SPF级Wistar大鼠随机分为两组:双眼形觉剥夺组与正常对照组。两组均在正常光照环境下饲养(12 h白天/黑夜交替环境),于视觉发育关键期后期(生后38-44 d)利用眼电生理诊断系统观察比较两组大鼠图形视觉诱发电位(pattern visual evoked potential,PVEP)中P100波潜时及振幅的差异,利用全细胞膜片钳技术记录两组大鼠视皮层单个神经元固有膜特性值的差异。结果双眼形觉剥夺组PVEP中P100波的潜时值为(95.67±14.67)ms,与正常对照组的(83.66±10.82)ms相比明显延长(P=0.002);双眼形觉剥夺组PVEP中P100的振幅为(3.02±1.02)μV,明显低于正常对照组的(7.42±1.65)μV,差异有统计学意义(P=0.000)。双眼形觉剥夺组与正常对照组视皮层神经元细胞的膜电容分别为(50.01±15.86)pF和(45.18±19.09)pF,膜电阻分别为(58.85±14.15)MΩ和(55.84±12.03)MΩ,时间常数值分别为(43.60±12.09)ms和(37.52±10.84)ms,两组相比差异均无统计学意义(均为P〉0.05)。结论生后早期双眼形觉刺激不足对视觉传导通路信号传递有延迟作用,但对视皮层单个神经元细胞的固有电生理学特性无影响。Objective To investigate the effects of binocular form deprivation on the electrophysiological characteristics of the visual pathway of postnatal rats. Methods Fifty Wistar rats of 8 postnatal days were randomly divided into binocular form deprivation group and normal control group. In this two groups,all of rats were housed in the same condition with reversed 1 2 hours /1 2 hours light / dark cycles. The latency and amplitude of P1 0 0-wave in these two groups were recorded by pattern visual evoked potential technique in postnatal 3 8- 4 4 days when visual development was at the end of the critical period.Meanwhile,intrinsic membrane characteristics of single visual cortical neuron of rats in these two groups were also recorded through whole-cell patch clamp technique for comparative analysis. R esults The latency of P1 0 0-were in binocular form deprivation group( 95. 67 ± 14. 67) ms was longer than that in normal control group( 83. 66 ± 10. 82) ms significantly( P = 0. 0 0 2),while the peak amplitude of P1 0 0-were in binocular form deprivation group( 3. 0 2 ± 1. 0 2) μV was much lower than that in normal control group( 7.4 2 ± 1. 6 5) μV( P = 0. 0 0 0). The membrane capacitance of visual cortical neurons in binocular form deprivation group and normal control group were( 5 0. 0 1 ± 1 5. 8 6) p F and( 45. 18 ± 19. 09) p F,respectively,the membrane resistance were( 58. 85 ± 14. 15) MΩand( 5 5. 8 4 ± 1 2. 0 3) MΩ,respectively,the time constant were( 4 3. 6 0 ± 1 2. 0 9) ms and( 3 7. 5 2 ± 1 0. 8 4) ms,respectively,there was no statistical difference between two groups( all P〉 0. 0 5). C onclusion The postnatal deficiency of binocular form leads to delayed signal induction of visual pathway,but it has no effect on the intrinsic electrophysiological characteristics of visual cortical neurons.
