摘  要:目的:探讨乌梅丸及其拆方对溃疡性结肠炎(UC)大鼠炎性因子及TLR9/My D88/NF-κBp65信号通路的影响,并分析其作用机制。方法:大鼠随机分为正常组、模型组、柳氮磺吡啶组、乌梅丸组、乌梅丸辛开苦降组、乌梅丸辛开组、乌梅丸苦降组、乌梅丸补益组。三硝基苯磺酸/乙醇法造模2d后给药4周,观察大鼠疾病活动指数(DAI)及结肠黏膜损伤程度,ELISA法检测结肠黏膜白介素-4(IL-4)、干扰素-γ(INF-γ)水平,免疫组化法观察Toll样受体9(TLR9)、髓样分化因子88(My D88)、细胞核因子κBp65(NF-κBp65)的表达。结果:乌梅丸组、乌梅丸辛开苦降组均能显著升高IL-4,降低INF-γ水平,显著减少结肠黏膜TLR9、My D88、NF-κBp65的蛋白表达(P<0.01),且效果优于其它乌梅丸拆方组。结论:乌梅丸组、乌梅丸辛开苦降组对UC大鼠结肠黏膜炎症有较好的缓解作用,其作用机制可能与调节炎性因子平衡,抑制TLR9/My D88/NF-κBp65信号通路的异常活化有关;辛开苦降、寒温并施的配伍方式可能是乌梅丸治疗UC的内在机制之一。Objective: To effects of Wumei Pill and its separated prescriptions on inflammatory factors and TLR9/My D88/NF-κBp65 signaling pathway in ulcerative colitis(UC) rat. Methods: Rats were randomly divided into normal group, model group, sulfasalazine group, Wumei Pill group, Wumei Pill-Xinkai Kujiang group, Wumei Pill-Xinkai group, Wumei Pill-Kujiang group, and Wumei Pill-Buyi group. The rat model of ulcerative colitis was induced by trinitrobenzen-sulphonic acid/ ethanol. After modelling for 2 days, rats were given the drug administration for 4 weeks. Then, disease activity index(DAI) and colonic mucosa damage were observed. The proteins expression of TLR9, My D88 and NF-κBp65 were detected by immunohistochemistry, and the levels of IL-4 and INF-α in the colonic mucosa tissue were detected by ELISA assay. Results: Wumei Pill group and its separated prescription groups could significantly increase the level of IL-4, and decrease the level of INF-γ and the proteins expression of TLR9, My D88 and NF-κBp65 in colonic mucosal tissue(P〈0.01). These effects in Wumei Pill group and Wumei Pill-Xinkai Kujiang group were superior to those in the other separated prescription groups. Conclusion: Wumei Pill and its Xinkai Kujiang separated prescriptions could improve the inflammation in the colonic mucosal tissue of UC, which may relate to regulate the balance of inflammatory factors and inhibit the abnormal activation of TLR9/My D88/NF-κBp65 signaling pathway. The compatibility of pungent drugs for dispersion, bitter drugs for purgation, cold property of drug, and warm property drug may be one of mechanism underlying the Wumei Pill in treating ulcerative colitis.
