摘  要:目的研究Nanog促进乳腺癌细胞侵袭与ezrinT567磷酸化的关系及Nanog调节ezrinT567磷酸化的可能机制。方法转染靶向Nanog的si RNA敲低乳腺癌细胞Nanog;用transwell法检测乳腺癌细胞侵袭;用免疫印迹检测乳腺癌细胞Nanog、PKCε表达水平和ezrinT567磷酸化水平;用免疫共沉淀和免疫印迹检测PKCε和ezrin蛋白的相互作用。结果敲低乳腺癌细胞的Nanog,PKCε蛋白表达下降、ezrinT567磷酸化水平下降、乳腺癌细胞的侵袭能力下降;敲低PKCε降低ezrinT567磷酸化水平;PKCε与ezrin可免疫共沉淀。结论 Nanog可上调PKCε的表达,后者可使ezrinT567磷酸化,这可能是Nanog促进肿瘤转移的一个新机制。Objective To study the relationship between Nanog- promoted metastasis of breast cancer and ezrinT567 phosphorylation, and explore the possible mechanism by which Nanog regulates ezrinT567 phosphorylation. Methods A siRNA construct targeting Nanog was transfected in breast cancer cells to knock down Nanog expression, and the changes in the cell invasion was detected using Transwell assay. The expression levels of Nanog and PKC and the phosphorylation level of ezrinT567 were detected using Western blotting and immunofluorescent staining; the protein interaction between PKC? and ezrin was assayed by co-immunoprecipitation and Western blotting. Results Nanog knockdown significantly decreased the expression of PKC-protein, phosphorylation level of ezrinT567 and the invasion ability of breast cancer cells. PKCεknockdown obviously decreased the phosphorylation level of ezrinT567 in the cells, and PKC? and ezrin were co-immunoprecipitated. Concludions Nanogcan can upregulate the expression of PKC? to promote the phosphorylation of ezrinT567, which can be a new mechanism by which Nanog promotes tumor metastasis.
