摘  要:目的探讨人参皂苷Rg1对脂多糖(LPS)诱导的BV2小胶质细胞炎症反应的作用及糖皮质激素受体(GR)阻断剂RU486的阻断效应。方法用LPS诱导BV2小胶质细胞炎症反应;在无或有RU486预处理细胞的情况下,相继用Rg1和LPS作用细胞。采用免疫印迹法检测诱导型一氧化氮合酶(iNOS)及环氧化酶2(COX2)水平的变化。结果 LPS可显著上调iNOS和COX2蛋白表达(F=13.42、29.74,q=7.571、15.320,P<0.01),Rg1预保护可明显降低由LPS诱导的iNOS和COX2蛋白表达上调(q=4.889、9.622,P<0.01),此作用可以被RU486所阻断(q=5.905、4.793,P<0.05)。结论人参皂苷Rg1能够抑制LPS诱导的BV2小胶质细胞iNOS和COX2蛋白表达,此作用可以被GR阻断剂RU486所阻断。Objective To investigate the effect of ginsenoside Rgl on lipopolysaccharide (LPS)-induced BV2 microglial inflammation and the blocking effect of glucocorticoid receptor (GR) antagonist RU486. Methods LPS was used to induce in- flammatory response in BV2 microglial cells. BV2 cells were treated with Rgl and LPS, one after another, in the absence or presence of GR antagonist RU486. Applying western blot, the changes of expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase 2 (COX2) were measured. Results LPS could significantly upregulate the protein expressions of iNOS and COX2 (F:13.42,29.74 q=7.571,15.320;P〈0.01). Rgl pretreatment could markedly decrease the upregulation of expressions of iNOS and COX2 induced by LPS (q=4.889,9.622;P〈0.01). These effects could be blocked by GR antagonist RU486 (q=5.905, 4.793;P〈0.05). Conclusion Rgl can inhibit expressions of iNOS and COX2 in BV2 microglial cells induced by lipopolysaccharide, and this action can be blocked by GR antagonist RU486.
