摘  要:目的:探讨卵巢癌SKOV3细胞凋亡的ERK1/2通路介导机制及白藜芦醇的参与作用。方法:将SKOV3细胞株经传代培养后随机分为5组,即对照组,白藜芦醇低、中、高剂量组和PD98059组。白藜芦醇低、中、高剂量组分别给予不同剂量白藜芦醇(5、50和500μmol·L^(-1))培养12 h,PD98059组在高剂量白藜芦醇培养后6 h加入PD98059(10μmol·L-1)培养。MTT法分析各组细胞的抑制率,Real-time PCR和Western blotting分析各组细胞中ERK信号通路及细胞凋亡相关蛋白的表达。结果:SKOV3细胞增殖抑制率及细胞凋亡蛋白表达随白藜芦醇浓度增高而升高或增强,且ERK信号通路Ras、Raf、ERK1/2及MEK蛋白的表达也逐步增强。PD98059组细胞的上述异常得到部分恢复。结论:白藜芦醇通过激活ERK信号通路发挥抑制SKOV3细胞增殖和促进凋亡的作用。Objective: To explore the involvement of ERK signaling pathway in resveratrol induced SKOV3apoptosis. Methods: SKOV3 cell lines were divided into 5 groups, i. e. , control group, low, middle and high dose of resveratrol groups and PD98059 group. Resveratrol group received resveratrol medication (5,50 and500 μmol · L-1) for 12 h while PD98059 group received PD98059( 10 μmol · L-1 )at the last 6 h. The inhibition rate and expression of ERK signaling pathway key proteins were detected. Results: The inhibition rate andCaspase3 expression were increased in resveratrol groups with the concentration of resveratrol, so was the expression of Ras, Raf, MEK and ERK. These parameters were normalized in PD98059 group to some extent. Conclusion:Resveratrol inhibits cell proliferation and promotes apoptosis by activating ERK signaling pathway in SKOV3 cell line.
