摘 要:本文研究了培养条件对海豚链球菌(Streptococcus iniae)NUF849生长的影响,分析了不同培养条件下海豚链球菌胞外产物的蛋白组成变化。结果显示,海豚链球菌在28℃时生长状态最好,胞外产物最丰富,主要有92 kDa、88 kDa、68 kDa、62 kDa、57 kDa、50 kDa、46 kDa、42 kDa、38 kDa、37 kDa、36 kDa、30 kDa、27 kDa和24 kDa等蛋白条带,温度升高或降低皆使蛋白条带减少;相对于普通气体环境,5%~10%CO_2对海豚链球菌的生长无显著影响,但使胞外产物蛋白条带减少,92 kDa、88 kDa、62 kDa、37 kDa和27 kDa条带丰度升高;培养基含2‰葡萄糖和5‰NaCl时,海豚链球菌生物量最高,5‰和10‰葡萄糖均可增加胞外产物蛋白条带,提高68 kDa、57 kDa、37 kDa和27 kDa蛋白丰度,但20‰葡萄糖和高于10‰的NaCl会抑制海豚链球菌的生长并减少胞外产物蛋白丰度;培养基中添加二联吡啶(DPD)和FeCl_3时,海豚链球菌生长受到抑制,胞外产物蛋白条带较BHI培养基条件下明显减少。本文结果有助于认识海豚链球菌对外界环境条件的反应,为揭示其致病机理提供依据。In this paper,the growth of Streptococcus iniae NUF849 under different culture conditions were studied,and the extracellular products(ECPs)of S.iniae were obtained using plate culture technique with their protein compositions profiled through SDS-PAGE.The results showed that the optimal growth of S.iniae and protein excretion of ECPs were detected under 28℃,and the bands of ECPs mainly included 92 kDa,88 kDa,68 kDa,62 kDa,57 kDa,50 kDa,46 kDa,42 kDa,38 kDa,37 kDa,36 kDa,30 kDa,27 kDa and 24 kDa protein,while some protein abundance were reduced with the temperature increasing or decreasing;Compared to ordinary gaseous environment,there was no significant effect on the growth of S.iniae with 5%~10% CO_2,but the protein bands of ECPs was reduced,although the abundance of several bands with molecular weight of 92 kDa,88 kDa,62 kDa,37 kDa and 27 kDa increased;The maximal biomass could be got when the medium contained 2‰ glucose and 5‰ NaCl,and the bands of ECPs were enriched at 5‰ and 10‰ of glucose with the protein abundance of 68 kDa,57 kDa,37 kDa and 27 kDa.However,20‰ of glucose and over 10‰ of NaCl inhibited the growth of S.iniae and the secretion of ECPs proteins.In addition,the growth of S.iniae was obviously inhibited and the bands of ECPs proteins were significantly decreased when DPD or FeCl_3 was added to the mediumas compared with BHI medium.The results revealed that the temperature,gaseous environment,salinity,glucose content and iron content in the medium had important effects on the growth of S.iniae and protein excretion of its ECPs,which would help us to have better understanding of responds of S.iniae to the condition changes and its pathogenesis.
