摘  要:目的:观察祛湿泄浊合剂对高尿酸血症模型小鼠尿酸、肾功能以及肝脏的黄嘌呤氧化酶(xanthine oxidase,XOD)、腺苷脱氨酶(adenosine deaminase,ADA)的影响。方法:采用氧嗪酸钾混悬液按照400 mg·kg-1的剂量一次性腹腔注射制备小鼠高尿酸血症模型,将84只小鼠随机分为对照组、模型组、痛风定对照组、祛湿泄浊合剂低剂量组、中剂量组、高剂量组。自由饮食7 d,使用氧嗪酸钾混悬液腹腔注射,2 h后取各组小鼠,摘眼球取血测定尿酸、肾功能。24小时后取小鼠肝脏,进行均浆,测定肝组织中XOD、ADA的活性。结果:与模型组比较,祛湿泄浊合剂中剂量组能降低氧嗪酸钾盐所致的小鼠高血尿酸,差异有统计学意义(P<0.05),与对照组比较,模型组能提高小鼠血尿酸水平,差异有统计学意义(P<0.05)。与模型组比较,其余各组小鼠的黄嘌呤氧化酶及腺苷脱氨酶的活性均增高,差异具有统计学意义(P<0.01);与对照组比较,模型组、祛湿泄浊合剂各剂量组黄嘌呤氧化酶及腺苷脱氨酶的活性明显改善,差异具有统计学意义(P<0.01)。结论:祛湿泄浊合剂能够有效降低高尿酸血症小鼠模型的血尿酸水平,可能与影响XOD、ADA酶的活性有关。Objective:To observe the clinical curative effect of Qushi Xiezhuo Mixture on Serum Uric Acid, kidney function and xanthine oridase (XOD) and adenosine deaminase (ADA) of the liver of model rats with hyperuricemia. Methods : The rat model with hyperuricemia was established by injecting potassium oxonate suspension into the abdominal cavity according to 400mg/kg standard each time. Then 84 rats were randomly divided into the control group, the model group, the Tongfeng Ding(TFD) control group,the group of low-dose Qushi Xiezhuo Mixture, the group of moderate-dose Qushi Xiezhuo Mixture, and the group of high- dose Qushi Xiezhuo Mixture. The rats took free diet for 7 days, then their abdominal cavity were injected into potassium oxonate suspension. Two hours later,the rats' eyeballs in each group were extirpated and their blood was taken to measure uric acid and renal function. Then 24 hours their livers were extirpated to measure XOD, ADA activity in liver tissue. Results : Compared with the model group, the group of moderate-dose Qushi Xiezhuo Mixture could reduce rat hyperuricemia due to potassium oxonate; And the difference between them had statistical significance(P 〈 0.05 ). Compared with the control group,the model group could im- prove rats' blood uric acid level;And the difference between them had statistical significance( P 〈 0.05 ). Compared with the mod- el group, Xanthine oxidase and adenosine deaminase activities of rats in the other groups all increased;And 'all the differences be- tween them had statistical significance(P 〈 0.01 ). Compared with the control group, Xanthine oxidase and adenosine deaminase activities of rats in the other groups all improved;And all the differences between them had statistical significance ( P 〈 0.01 ). Conclusion:Qushi Xiezhuo Mixture can effeetively reduce blood uric acid level of model rats with hyperuricemia,which probably has something to do with activities of XOD and ADA.
