维吾尔族结直肠癌miRNA基因表达与疾病预后关系的研究

摘　　要：目的:探讨维吾尔族结直肠癌组织中差异性表达的miRNA及其与结直肠癌预后的关系。方法:收集手术切除的维吾尔族结直肠癌患者结直肠癌组织和维吾尔族正常结直肠黏膜组织标本,应用AFFX miRNA表达谱芯片实验初步筛查癌及正常组织中miRNA的差异表达,并采用实时定量PCR技术验证芯片检测结果,分析差异性表达的miRNA与维吾尔族结直肠癌临床病理学关系。结果:30种miRNA在维吾尔族结直肠癌患者切除癌组织与接受痔痛手术患者的结直肠正常黏膜组织相比存在显著差异表达(<0.05)。18个表达上调的miRNAs分别为hsa-miR-31,hsa-miR-196b,hsa-miR-105,hsa-miR-224,hsa-miR-27b,hsa-miR-106a,hsa-miR-486-5p,hsa-miR-483-6p,hsa-miR-663b,hsa-miR-574-3p,hsa-miR-584,hsa-miR-378,hsa-miR-141,hsa-miR-18a,hsa-miR-19a,hs a-miR-452,hsa-miR-622和hsa-miR-768-5p,12个表达下调的分别是hsa-miR-215,hsa-miR-4298,hsa-miR-1246,hsa-miR-139-5p,hsa-miR-34a,hsa-miR-3201,hsa-miR-363,hsa-miR-133b,hsa-miR-133a,hsa-miR-378a-3p,hsa-miR-1825和hsa-miR-30a,其中以hsa-miR-196b,hsa-miR-31和hsa-miR-106a在结直肠癌组织中表达升高具有显著性。实时荧光定量PCR检测结果与miRN A芯片检查的结果基本吻合,证实miRNA芯片结果真实可靠。结论:成功筛选出维吾尔族结直肠癌组织中差异性表达的miRNA;hsa-miR-196b,hsa-miR-31和hsa-miR-106a的高表达与维吾尔族结直肠癌的转移有关,并提示疾病的预后不良。Objective： To investigate the miRNA expression profiling in colorectal carcinoma and analyze the differently expressed miRNAs.Methods： Uyghur nationality patients＇ colorectal carcinoma tissue and healthy tissue were collected and AFFX miRNA expression chip was used to determine the differently expressed miRNAs.The results were further verified by fluorescent quantitation polymerase chain reaction.Results：30 kinds of miRNAs were found to differently express in colorectal carcinoma tissue and healthy tissue.18 miRNAs was found up-regulated expression included hsa-miR-31,hsa-miR-196 b,hsa-miR-105,hsa-miR-224,hsa-miR-27 b,hsa-miR-106 a,hsa-miR-486-5p,hsa-miR-483-6p,hsa-miR-663 b,hsa-miR-574-3p,hsa-miR-584,hsa-miR-378,hsa-miR-141,hsa-miR-18 a,hsa-miR-19 a,hsa-miR-452,hsa-miR-622 and hsa-miR-768-5p.12 miRNAs was found down-regulated expression,include hsa-miR-215,hsa-miR-4298,hsa-miR-1246,hsa-miR-139-5p,hsa-miR-34 a,hsa-miR-3201,hsamiR-363,hsa-miR-133 b,hsa-miR-133 a,hsa-miR-378a-3p,hsa-miR-1825andhsa-miR-30 a.The highest up-regulated miRNAs was hsa-miR-196 b,hsa-miR-31 and hsa-miR-106 a.The results of fluorescent quantitation polymerase chain reaction correlated with that of miRNA expression chip,indicating a credible results.Conclusion： Differently expressed miRNAs were successfully detected,which could be used as auxiliary diagnosis biomarkers.hsa-miR-196 b,hsa-miR-31 and hsa-miR-106 a level was positively correlated with colorectal carcinoma and indicated a bad prognosis.
