摘  要:目的建立一种测定全蝎中未衍生化牛磺酸含量的方法。方法以高效液相色谱-蒸发光散射检测器(HPLC-ELSD)法,采用Prevail C18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.7%三氟乙酸为流动相进行洗脱,洗脱时间为15 min。结果牛磺酸峰面积的自然对数与相应浓度的自然对数呈良好的线性关系,其方程为Y=1.4475X+4.0088,R^2=0.9998,线性范围为0.05~0.5 mg·mL^(-1),平均回收率为102.77%,RSD为2.25%,4批全蝎牛磺酸含量为0.81%~2.02%。结论该法快速、简便、准确,可作为全蝎药材的质量控制方法。Objective To establish a content determination method for pre- column derivatization taurine in scorpion.Methods High performance liquid chromatography(HPLC)- evaporative light- scattering detector(HPLC- ELSD)method was used for the detection. The determination was performed on Prevail C18(4.6 mm×250 mm, 5 μm)column, with methanol- 0.7 % trifluoroacetic acid as mobile phase and elution time being 15 minutes. Results The natural logarithm of peak area and the natural logarithm of corresponding concentration of taurine had good linear relationship, the equation was Y=1.4475X+4.0088, R2=0.9998, and the linear range was 0.05~0.5 mg·m L^(-1), the average recovery was 102.77%,and RSD was 2.25 %. The contents of taurine were from 0.81 to 2.02 % in scorpio of 4batches. Conclusion The method is simple,rapid and accurate,and can be used for quality control of scorpion.
