摘 要:采用高效液相色谱-蒸发光散射检测法测定木薯中果糖、葡萄糖、蔗糖和海藻糖的含量。样品用水经超声波提取,用Xbridge-NH_2色谱柱分离,流动相为乙腈∶水(水中加入0.2%的三乙胺)的体积比为70:30,流速为1.0m L/min,Elsd漂移管温度为85℃,氮气流速2 L/min。在该条件下果糖、葡萄糖、蔗糖和海藻糖得到很好的分离,分别在1.5~25,1.5~25,2~25和1.5~25μg的范围内有良好的线性范围,相关系数R^2分别为0.999 1,0.999 1,0.999 0和0.999 3,最低检出限分别为6.50,5.50,4.00和7.00 mg/kg,加标回收率分别为97.6%,98.6%,96.2%和95.4%。验证试验结果表明,该方法快速、准确,能用来测定木薯中单糖的含量。Monosaccharides in cassava samples were determined by HPLC-ELSD. The sample was extracted by water and separated on the Xbridge-NH_2 chromatographic column using acetonitrile/water(70/30) supplemented with 0.2% triethylamine as the mobile phase, and the flow rate of 1.0 m L/min. The ESLD drift tube temperature was set to 85 °C and the nitrogen flow velocity was 2 L/min. In these conditions, the fructose, glucose, sucrose and trehalose could be clearly separated. The fructose, glucose, sucrose and trehalose showed a good linearity in 1.5~25, 1.5~25 and 2~25 μg(R2=0.999 1, 0.999 1, 0.999 0 and 0.999 3), the limits of detectin(LOD) were 6.50, 5.50, 4.00 and 7.00 mg/kg and the recoveries for spiked samples were 97.6%, 98.6%, 96.2% and 95.4%. The verified results demonstrated that this method was precise and accurate and could be used for determination of monosaccharides in cassava.
