CDKN2A、CDKN2B、MEF2A、VEGF基因表达与冠状动脉粥样硬化性心脏病发生的相关性

摘  要:目的比较细胞周期蛋白依赖性激酶抑制因子2A(CDKN2A)、CDKN2B、肌肉细胞特异性增强因子2A(MEF2A)以及血管内皮生长因子(VEGF)基因表达在冠状动脉粥样硬化性心脏病(CHD)患者和健康人之间的表达差异,以期探寻CHD可能的发病机制。方法选取CHD患者154例(CHD组)和健康者123例(正常对照组),留取全部研究对象的血液标本。通过实时定量PCR法检测CDKN2A、CDKN2B、MEF2A和VEGF mRNA的表达;通过ELISA定量检测血清中CDKN2A、CDKN2B、MEF2A和VEGF的含量。结果实时定量PCR结果发现,CHD组CDKN2A、CDKN2B、MEF2A和VEGF mRNA表达量分别为0.011 5±0.002 1、0.003 3±0.000 4、0.302 6±0.035 6和0.022 1±0.004 2,均显著低于正常对照组(分别为0.048 4±0.015 6、0.007 1±0.001 2、0.823 0±0.075 1和0.052 7±0.001 3),差异均有统计学意义(均P<0.05)。ELISA检测结果表明,CHD组血清CDKN2A、CDKN2B、MEF2A及VEGF的表达量分别为(121.60±7.96)、(65.07±5.42)、(6.51±0.83)和(19.41±1.62)μg/mL,均显著低于正常对照组[分别为(236.90±29.13)、(155.70±22.52)、(15.96±2.86)和(39.87±4.54)μg/mL],差异均有统计学意义(均P<0.05)。结论 CHD患者血液中有核细胞内CDKN2A、CDKN2B、MEF2A和VEGF mRNA的表达水平以及血清中蛋白的表达水平都显著低于健康人群,提示这4个基因可能直接影响或间接参与CHD的发生、发展。Objective To explore the relationship between coronary heart disease(CHD)and expressions of cyclin-depedent kinase inhibitor 2A(CDK2A),CDK2B,myocyte enhancer factor 2A(MEF2A)and vascular endothelial growth factor(VEGF)genes. Methods A total of 154 CHD patients(CHD group)and 123 healthy control cases(control group)were enrolled in the study. All blood samples from patients and control cases were centrifuged for serum collection,and the RNA was extracted after red cell lysis. The gene expressions and protein levels were detected by qualitative real-time PCR(q RT-PCR)and enzyme-linked immunosorbent assay(ELSIA). Results Expressions of CDKN2A,CDKN2B,MEF2A and VEGF mRNA(0.011 5±0.002 1,0.003 3±0.000 4,0.302 6±0.035 6 and 0.022 1±0.004 2,respectively)in blood cells of CHD patients were significantly lower than those of control cases(0.048 4±0.0156,0.007 1±0.001 2,0.823 0±0.075 1 and 0.052 7±0.001 3,P〈0.05). Meanwhile,serum concentrations of CDKN2A,CDKN2B,MEF2A and VEGF(121.60±7.96,65.07±5.42,6.51±0.83 and 19.41±1.62 μg/mL,respectively)of CHD patients were also lower than those of control cases(236.90±29.13,155.70±22.52,15.96±2.86 and 39.87±4.54 μg/mL,P〈0.05). Conclusion CHD patients had lower expression of CDKN2A,CDKN2B,MEF2A and VEGF than healthy people.
