摘　　要：目的探讨脂联素(Apn)对骨髓间充质干细胞(MSCs)的增殖及凋亡的影响。方法分离培养原代大鼠MSCs,用构建的小鼠Apn基因重组慢病毒(Lenti-Apn-IRES-EGFP)或对照空载病毒(Lenti-IRES-EGFP)转染MSCs,并测定转染率。转染后用CCK8试剂盒检测Apn-MSCs、Gfp-MSCs在体外的增殖情况,并在缺氧缺糖损伤模型(OGD)12 h后用流式细胞仪检测细胞凋亡率。结果与对照组比较,Apn-MSCs细胞增殖速度更快(P=0.001)。在OGD 12 h后,Apn-MSCs凋亡率比对照组低(P=0.01)。结论 Apn可以促进MSCs的增殖,并减少缺氧、缺糖损伤后MSCs的凋亡。Objective To explore the effects of adiponectin（ Apn） on the proliferation and apoptosis of m esenchym al stem cells（ MSCs）. Methods Prim ary rat M SC s were separated and cultured. MSCs were transfected with constructed m ouse Apn gene recom binant lentivirus（ Lenti-Apn-IRES-EG FP） or control lentivirus（ Lenti-IRES-EG FP） and the transfection efficiency was m easured. The in vitro proliferation of G fpM SC s and Apn-M SC s were detected w ith the C C K8 kit after transfection. The apoptosis rate was m easured with the flow cytom eter 12 h after the construction of oxygen and glucose deprivation（ O G D） m odel. Results The proliferation of Apn-M SC s w as faster as com pared with G fp-MSCs（ P = 0. 001）. The apoptosis rate of Apn-M SC s was lower than that of G fp-M SC s 12h after the construction of O G D m odel（ P = 0. 01）.Conclusion Apn can prom ote the proliferation of MSCs and reduce the apoptosis of M SC s after oxygen and glucose deprivation.
