摘 要:目的:借鉴基因芯片技术,以性味药理学理论为指导,探讨薏苡仁不同组分对利湿作用的主要机制。方法:以薏苡仁不同拆分组分对脾虚水湿模型大鼠进行治疗,对空肠组织进行全基因表达谱检测,对GO分子功能的Transporter activity功能注释到的基因进行主成分分析和聚类分析,比较各治疗组疗效和可能机制。结果:薏苡仁蛋白、多糖和脂肪油对脾虚水湿大鼠Transporter activity功能具有明显调节作用;而相关基因团以Slc26a、Slc5a、Slc13a、AQP3、AQP7、Fabp6等为主;它们对转运活性基因调节具有共性,可能具有共同的作用位点和作用途径;淀粉对脾虚水湿模型大鼠Transporter activity功能改善较弱。结论:薏苡仁调节健脾利湿大鼠模型转运功能的物质可能存在于蛋白、多糖和脂肪油中,淀粉(残渣)中健脾利湿的功能较弱。Objective: Based on genomics and under the guidance of TCM five- flavor theory to discuss the influence and possible mechanism of adlay different components working on the jejunum transporting activity of rats of spleen deficiency with water retention. Methods: The rats of spleen deficiency with water retention were treated with adlay different components and we examined the expressions of the entire genes of jejunum tissue. After obtaining the genes annotated by the transporter activity function of GO,we executed the main component analysis and the classification analysis and compared the effect of each treatment group and the possible mechanism. Results: The protein,polysaccharide and oil of adlay had obvious regulation effect on the rats of spleen deficiency with water retention. While the similar gene clusters were mainly Slc26 a,Slc5a,Slc13 a,AQP3,AQP7 and Fabp6,which shared the same nature in transporting active genes regulation and may share the same effect position and route. The function of starch working on the rat model of spleen deficiency with water retention was weak. Conclusion: The main material basis of adlay which can regulate the transporting function of the rat model of tonifying spleen and removing dampness may exist in protein,polysaccharide and oil,and such function of starch( residue) is comparatively weak.
