摘 要:目的观察消渴通痹颗粒对糖尿病大鼠下肢缺血肌组织缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)mRNA表达的影响。方法取SD大鼠60只,随机取10只作为空白组,其余大鼠采用高脂饮食复合链脲佐菌素、股动脉结扎法建立糖尿病大鼠下肢缺血模型。将造模成功大鼠随机分为模型组,消渴通痹颗粒大、中、小剂量组及西医药组,空白组及模型组给予生理盐水10 m L/(kg·d)灌胃,消渴通痹颗粒大、中、小剂量组分别给予消渴通痹颗粒1.875 g/(kg·d)、1.35 g/(kg·d)、0.625 g/(kg·d)灌胃,西药组给予西洛他唑45 mg/(kg·d)灌胃,连续灌胃6周。观察大鼠一般情况及体质量、血糖变化,取缺血侧肌肉采用实时定量PCR法检测HIF-1αmRNA和VEGFmRNA表达情况。结果消渴通痹颗粒大、中、小剂量组灌胃6周后大鼠体质量均明显高于模型组(P均<0.05),且消渴通痹颗粒中剂量组大鼠体质量明显高于消渴通痹颗粒大、小剂量组(P均<0.05);消渴通痹颗粒中剂量组血糖水平明显低于模型组及其他给药组(P均<0.05)。模型组HIF-1αmRNA和VEGFmRNA表达量均明显高于空白组(P均<0.05),各给药组HIF-1αmRNA和VEGFmRNA表达量均明显低于模型组(P均<0.05),且消渴通痹颗粒中剂量组HIF-1αmRNA和VEGFmRNA表达量均明显低于消渴通痹颗粒大剂量组(P均<0.05)。结论消渴通痹颗粒可通过增加缺血部位肌肉组织中HIF-1αmRNA和VEGFmRNA表达而促进缺血组织新生血管形成,改善糖尿病肢体缺血状态。Objective It is to observe the effects of Xiaoke Tongbi Granule on the expression of hypoxia inducible factor-1α( HIF-1α) and vascular endothelial growth factor( VEGF) in ischemia tissue of lower limb muscle of diabetic rats.Methods 10 SD rats were randomly selected as the blank control group,and the other rats were established by high fat diet,STZ and femoral artery ligation was used to establish the model of the lower limb ischemia in diabetic rats. successful model rats were randomly divided into model group,large,medium and small dose groups for Xiaoke tongbi granule,and western medicine group,blank control group and model group were given normal saline 10 m L /( kg·d),Xiao Ke Tongbi Granules of large,medium and small dose groups were treated with Xiaoke Tongbi Granule 1. 875 g /( kg·d),1. 35 g /( kg·d),0. 625 g /( kg·d) by gavage. The western medicine group was given cilostazol 45 mg /( kg·d) by gavage,all the rats were treated for 6 weeks. The general situation,body weight and blood glucose of rats were observed,and the expression of HIF-1mRNA and VEGFmRNA were detected by real-time quantitative PCR. Results The weight of the rats of large,medium and small dose group were significantly higher than those in the model group( P〈0. 05),after 6 weeks' treatment. Moreover,the rats body weight of medium dose group was significantly higher than that of large and small dose group( P〈0. 05). Blood sugar level of medium dose group was significantly lower than that of model group and other groups( P〈0. 05). In model group the expression levels of HIF-1αmRNA and VEGFmRNA were significantly higher than those in the blank group( P〈0. 05); in the treatment group the expression levels of HIF-1αmRNA and VEGFmRNA were significantly lower than those in the model group( P〈0. 05); the expression levels of HIF-1αmRNA and VEGFmRNA in medium dose group were significantly lower than that of large dose group( P〈0. 05).
