摘 要:目的研究血根碱(Sanguinaria canadensis,SAN)诱导人乳腺癌细胞MCF-7凋亡可能的作用机制。方法 MTT法检测SAN对MCF-7细胞存活率的影响;激光扫描共聚焦显微镜观察SAN对MCF-7细胞内活性氧(reactive oxygen species,ROS)含量的影响;化学发光法检测SAN对MCF-7细胞内caspase-3、caspase-8、caspase-9活性的影响。结果 SAN能降低MCF-7细胞存活率;SAN可明显升高MCF-7细胞内ROS含量,升高细胞内caspase-3、caspase-8、caspase-9的活性,而这种作用能被抗氧化剂NAC抑制。结论以上结果证明SAN能够抑制人乳腺癌细胞MCF-7增殖,其机制可能是通过升高细胞内ROS水平,激活caspase级联反应,进而诱导细胞凋亡发挥作用。Aim To investigate whether exposure to Sanguinarine( SAN) can inhibit cell proliferation in human mammary adenocarcinoma cells( MCF-7) and the possible mechanism. Methods We exposed MCF-7 to anticancer compound SAN,cell viability was assessed by using the 3-( 4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide( MTT) reduction assay.ROS was measured using confocal microscopy,expression of caspase-3,caspase-8 and caspase-9 were calculated using chemiluminescence method. Results SAN remarkably inhibited growth of human mammary adenocarcinoma MCF-7 cells by decreasing cell proliferation.ROS release and caspase-3,caspase-8,caspase-9 expression were stimulated by SAN in MCF-7,and these changes were abolished by the antioxidant,N-acetylcysteine( NAC). Conclusion Regulation of caspases expression and release from MCF-7 cells are possibly evoked by SAN through reactive oxygen species.
