实验性牙周炎大鼠牙周组织、龈沟液及外周血中IL-22的检测

摘　　要：目的:在大鼠实验性牙周炎模型上检测炎症性牙周组织、龈沟液、外周血中白细胞介素22(IL-22)及其受体(IL-22R1)的表达水平。方法:取16只SD大鼠随机分为2组(n=8),实验组采用丝线结扎及牙龈卟啉单胞菌液涂抹法建立牙周炎模型;对照组无任何干预措施。8周后采集外周血、龈沟液及颌骨样本,体视显微镜及光镜下观察牙周组织病理改变,免疫组化分析牙周组织中IL-22、IL-22R1的表达水平,实时荧光定量PCR检测牙周组织中IL-22、IL-22R1 mRNA的表达水平,酶联免疫吸附法检测外周血及龈沟液中IL-22的水平。结果:实验组大鼠牙周组织病理改变符合牙周炎病理改变特征,牙槽骨吸收显著高于对照组(P<0.05);牙周组织中IL-22、IL-22R1及其mRNA表达较对照组显著上调(P<0.05);龈沟液及外周血清中IL-22浓度均较对照组显著升高(P<0.05)。结论:IL-22参与牙周炎免疫反应。AIM： To detect the expression of interleukin-22（ IL- 22） and IL-22 receptor（ IL-22R1） in periodontal tissues, gingival crevicular fluid and peripheral blood of SD rats with experimental periodontitis.METHODS： 16 SD rats were randomly divided into control group（ n = 8） and periodontitis group（ n = 8） in which experimental periodontitis was initiated by silk thread ligation and daubing bacteria. Periodontal tissues,gingival crevicular fluid and peripheral blood of each group were sampled after 8 weeks. The periodontal tissues were observed by HE staining. IL-22 and IL-22R1 expression in periodontal tissues was examined by immunohistochemistry and real- time quantitative PCR. ELISA was used to detect the concentration of IL- 22 in gingival crevicular fluid and peripheral blood. RESULTS： Periodontal inflammation and alveolar bone absorption were observed in the reats of periodontitis group. The protein and mRNA expression of IL-22 and IL-22R1 in the inflamed periodontal tissues was higher than in the controls（ P〈0. 05）. The concentrations of IL-22 in gingival crevicular fluid and peripheral blood of periodontitis group were higher than those of control group（ P〈0. 05）. CONCLUSION： IL-22 may be associated with the progression of periodontitis.
