摘 要:目的探讨丙酮酸钠(SP)对红细胞储存损伤的影响。方法戊巴比妥钠腹腔注射麻醉小鼠,同时机械通气,通过心脏穿刺取血,CPDA-1作为抗凝剂和储存液,过滤去除白细胞后平分2组(n=8),SP组:添加SP浓缩液使储存红细胞中SP的终浓度为2.5 mmol/L;生理盐水(NS)组:按相同体积比加入等量的生理盐水。400 g离心15min,调整Hct为70%-75%,4℃储存14 d后,用全自动血细胞计数仪测定储存红细胞的血常规;分别用三磷酸腺苷(ATP)、丙二醛(MDA)测定试剂盒测定储存红细胞中ATP、MDA水平。结果储存14 d时,2组血细胞计数各指标无明显差异(P>0.05),且均在正常范围。NS与SP组红细胞中ATP含量(nmol/mg)为:2.46±0.19 vs 2.82±0.21(P<0.05);MDA含量(nmol/mg)为15.45±0.33 vs 14.66±0.87(P<0.05)。结论在红细胞储存过程中,SP明显提高了储存红细胞ATP水平、降低了MDA水平,具有改善红细胞储存损伤的作用。Objective To investigate the effect of sodium pyruvate( SP) on red blood cells( RBCs) damage during storage. Methods The mice were anesthetized with intraperitoneal injections of sodium pentobarbital and mechanically ventilated. Mice were bled by cardiac puncture into CPDA-1 for anticoagulant and storage solution. The whole blood was leu-koreduced and divided into two groups equally( n = 8) : 1). SP group,where the concentrated solution of SP was added to RBC suspension to make the SP concentration 2. 5 mmol / L in a RBCs storage solution; 2) normal saline( NS) group,where RBCs suspension was added to saline at the same ratio as SP group,then was centrifuged at 400 g for 15 min. Its volume was reduced to a final hematocrit of 70% to 75%. The RBCs were stored at 4℃ for 14 days. After that,red blood cell counts were measured by blood cell automatic counter. The ATP level and MDA level were measured by ATP and MDA assay kits,respectively. Results After 14 days of storage,red blood cell counts in both of the two groups had no significant difference( P 0. 05),and all the indexes are normal. The ATP content of RBCs in NS group and SP group( nmol / mg protein) were 2. 46 ± 0. 19 vs 2. 82 ± 0. 21( P 0. 05). The MDA content in NS group and SP group( nmol / mg protein) were 15. 45 ± 0. 33 vs 14. 66 ± 0. 87( P 0. 05). Conclusion During RBC storage,according to the results of the ATP levels and MDA levels in two groups,the SP plays an important role in alleviating the lesion that occurred during storage.
