摘  要:目的探讨下调Snail基因对子宫内膜癌增殖和侵袭的影响及其机制。方法培养子宫内膜癌细胞株,构建Snail-shRNA表达载体,建立下调Snail基因表达的子宫内膜癌细胞株;用CCK-8法检测子宫内膜癌细胞株(Ishikawa)的增殖能力。用Transwell小室结合基质胶(Matrigel)法比较子宫内膜癌细胞株与相应的瞬时转染细胞株在细胞迁移能力及侵袭能力的差异。结果通过下调Snail基因在细胞内的表达,体外观察子宫内膜癌细胞增殖能力空白对照组高于细胞转染组;迁移与侵袭能力空白对照组高于细胞转染组。结论 Snail基因在子宫内膜癌中表达下调;与抑制子宫内膜癌的增殖和侵袭转移相关,其可能机制与逆转肿瘤的上皮细胞-间质转化过程和共同抑制下游转移靶基因相关。Purpose To investigate the effect of down-regulation of Snail gene on proliferation and invasion of endometrial carcinoma and its mechanism. Methods The endometrial carcinoma cell lines was cuhured, expression vector of Snail-shRNA was constructed, and endometrial carcinoma cell lines with down-regulation of Snail gene expression was established. CCK-8 was used to determine endometrial carcinoma cell lines (Ishikawa) proliferation. Migration ability and invasion ability of endometrial carcinoma cell lines and corresponding transient transfection cell lines was compared with transwell chamber combined with the matrix (Matrigel). Results By blocking the expression of Snail gene in cells, in vitro exprement showed the proliferation of endometrial cancer cells in blank control group was higher than that of cells transfection group, migration and invasion ability in the blank control group was higher than that of cells transfeetion group. Conclusion Down-regulation of Snail gene expression in endometrial carcinoma may associate with the inhibition of endometrial carcinoma proliferation and invasion, its possible mechanism may be related to the inversion of epithelial-mesenchymal transition process of the tumor and co-suppression of the downstream metastatic target genes.
