摘 要:对生产上广泛应用的4个优质籼型杂交水稻保持系中9B、金23B、Ⅱ–32B和川香29B,2个恢复系9311和明恢63的组培特性进行系统研究,发现川香29B愈伤组织的诱导率、继代培养增殖率、耐继代能力及分化再生能力均高于其他籼稻品种,其愈伤诱导率和分化再生率分别为96%和82%,说明川香29B是1个具有优良组培特性的籼稻品种;进一步对川香29B的农杆菌遗传转化体系进行优化,并初步建立了川香29B稳定的农杆菌介导的转化体系,即农杆菌EHA105菌液浓度OD600 nm为0.1,浸染液AAM中乙酰丁香酮浓度为200μmol/L,浸染时间30 min,黑暗条件下共培养72 h,共培养温度为19℃,抗性植株转化率为19.6%。Characteristics of in vitro plant tissue culture of four widely applied elite Indica hybrid rice maintainer lines (Zhong9B, Jin23B, II-32B,Chuanxiang29B)and two widely applied elite restorer lines(9311 and Minghui63) were investigated systematically in this study. The Indica rice cultivar-Chuanxiang29B was identified with excellent tissue culture response. It is shown that the callus induction rate of Chuanxiang29B from mature embryos was 96% and the plantlet regeneration rate of it was 82%, indicating higher rates of callus induction, proliferation, subculture tolerance and plantlet regeneration of Chuanxiang29B than most of other indica rice cultivars. An optimized and stable genetic transformation system mediated by Agrobacterium tumefacien for Chuanxiang29B was then established, in which the transformation efficiency of resistant plants reached up to 19.6% with the following conditions: the period of the calli inoculation with Agrobacterium tumefacien EHA105 (09600 nm=0.1) in AAM with acetosyringone (200 μmol/L) for 30 min and cocultivation at 19 ℃ in dark for 72 h.
