摘　　要：目的研究白细胞介素(IL)-8及其受体(R)A和B mRNA在Hela宫颈癌细胞株的表达及外源性IL-8对Hela细胞增殖的作用。方法实时聚合酶链反应(RT-PCR)方法检测IL-8及IL-8RA和IL-8RB在Hela细胞的表达;MTT法检测不同浓度的外源性IL-8对Hela细胞增殖的影响;RT-PCR方法检测外源性IL-8对IL-8RA和IL-8RB表达的影响。结果 Hela细胞表达IL-8及IL-8RA和IL-8RB,并且IL-8RA的表达量明显高于IL-8RB;外源性IL-8可显著促进Hela细胞的增殖,并明显上调IL-8RA和IL-8RB的表达。结论外源性IL-8可通过上调Hela细胞IL-8RA和IL-8RB的表达而促进其增殖。Objective To investigate mRNA expressions of IL-8 and its receptors IL-8RA and IL-8RB in Hela uterine cervix cancer cell line and the effect of exogenous IL-8 on the proliferation of Hela cell.Methods The expressions of IL-8 and IL-8RA and IL-8RB were detected by RT-PCR assay; the influence of different concentrations of exogenous IL-8 on proliferation of Hela cell was detected by MTT assay; the effects of exogenous IL-8 on the expressions of IL-8RA and IL-8RB were detected by RT-PCR assay.Results IL-8 and IL-8RA and IL-8RB were expressed in Hela uterine cervix cancer cell line,and the expression level of IL-8RA was significantly more than that of IL-8RB; exogenous IL-8 significantly promoted proliferation of Hela cell and up-regulated the expression of IL-8RA and IL-8RB.Conclusions Exogenous IL-8 could up-regulate the expressions of IL-8RA and IL-8RB and ultimately promote proliferation of Hela cell.
