摘  要:以甘油三酯型鱼油为原料,利用天然DHA相对富集于甘油酯2位的特点,以1,3-特异性固定化脂肪酶催化制备Sn-2二十二碳六烯酸甘油单酯(Sn-2 DHA-MAG)。建立了"格氏反应-薄层色谱-高效液相色谱"联用以Sn-2位DHA含量为标准筛选最佳反应原料的方法,随后考察了反应体系,硼酸浓度,反应温度和反应时间等因素以优化反应条件。结果表明,以优选的甘油三酯型鱼油为原料,50 mg/m L硼酸乙醇溶液∶鱼油∶固定化脂肪酶Lipozyme TL IM质量比等于40∶80∶18构建反应体系,在常温下反应4 h,反应产物减压蒸馏后,经硅胶柱层析纯化,所得Sn-2二十二碳六烯酸甘油单酯的纯度达90%以上。Sn- 2 docosahexaenoic acid monoglyceride was synthesized by ethanolysis of triglyceride fish oil with1,3- specific immobilized lipase. Raw materials were screened according to the Sn- 2 DHA content of the triglyceride by a method combining Grignard reaction,TLC and HPLC. The influence of reaction system,boric acid concentration,reaction temperature and time were studied to optimize the conditions for the synthesis. Taken together,the reaction system consisted of 50 mg/m L boric acid ethanol solution: optimized fish oil: immobilized lipase Lipozyme TL IM with a 40∶ 80∶ 18 mass ratio,and the reaction was conducted at room temperature for 4 h. The product was distilled under reduced pressure and then purified by silica gel column chromatography. Under this condition,Sn- 2DHA- MAG of up to 90% purity was synthesized.
