摘  要:目的:筛选喉癌颈部淋巴结转移相关分子标记物。方法:纳入喉癌伴或不伴颈部淋巴结转移病例共30例,收集原发灶、切缘黏膜、颈部转移淋巴结标本,分为切缘组(n=30)、原发灶组(n=30)、淋巴结组(n=16),采用RTq PCR分别检测HGF、c-MET、VEGF、EGF、VASH1、HIF-1α、JMJD3、JMJD4基因m RNA表达,筛选颈部淋巴结转移相关基因。结果:HGF、VEGF、EGF、HIF-1α、JMJD3基因在喉癌组织中呈高表达(均P<0.05),VASH1则为低表达(P<0.05);进一步筛选表明,HIF-1α、JMJD3在颈部转移淋巴结中的表达明显高于原发灶(均P<0.05)。结论:HIF-1α、JMJD3过表达可能与颈部淋巴结转移有关,值得进一步研究。Objective To screen the molecular markers related to cervical lymph node metastasis in laryngeal cancer. Methods A total of 30 cases of laryngeal cancer with or without cervical lymph node metastasis were included, primary tumor foci was collected, edge of mucosa and neck metastatic lymph node were cut, and margin group(n = 30), the primary lesion group(n = 30), lymph node(n = 16) were designed. RT q PCR was used to detect the expression of HGF, c-Met, VEGF, EGF, VASH1, HIF-1α, JMJD3 and JMJD4 m RNA, cervical lymph node metastasis related genes were screened. Results HGF, EGF and VEGF, HIF-1αand JMJD 3 gene in laryngeal carcinoma tissues showed high expression(P 0.05), While VASH1 showed low expression(P 0.05); Further screening showed that HIF-1α and JMJD3 expression in cervical metastasis lymph nodes were significantly higher than that of the primary tumor foci(P 0.05). Conclusion The overexpression of HIF-1α and JMJD3 may be related to the metastasis of cervical lymph nodes, and it is worth further study.
