摘　　要：目的:检测长链非编码RNA(lncRNA)ANRIL在肾癌及正常组织、细胞中的表达水平,探究其在肾癌中的临床意义。方法:提取肾癌细胞(ACHN、786O和769P)、正常人胚肾细胞(293T)和47对组织中的总RNA,逆转录获取cDNA,之后通过qPCR方法检测细胞及组织中的ANRIL表达量,分析配对组织、细胞系之间的表达差异,探究ANRIL表达水平与患者临床特征之间的关系。结果:ANRIL在肾癌细胞和组织中表达水平明显低于正常肾细胞(P<0.05)和配对癌旁正常组织(P<0.01),标本中共有34例(72.34%)肾癌组织ANRIL表达低于癌旁组织,癌组织中ANRIL表达与患者年龄、性别及肾癌病理类型无明显相关性(P>0.05),与患者TNM分期和AJCC分级相关(P<0.05)。结论:ANRIL在肾癌组织及细胞系中表达明显下调,且和肾癌临床分级相关,提示其在肾癌发生发展中发挥着一定的作用。Objective：To detect the expression level of long non-coding RNA（lncRNA）ANRIL in paired tissues and cell lines,and explore ANRIL＇s clinical significance in renal cell carcinoma.Method：Total RNA was isolated from RCC cell lines（ACHN,786 O,769P）,human embryo kidney（293T）and 47 paired tissues,and then reverse transcription was performed to get cDNA.Real time quantitative PCR（RT-qPCR）was performed to detect the expression of ANRIL.Statistic analysis was performed to analyse the expression of ANRIL and explore the correlations between the expression level of ANRIL and patients＇ clinical characteristics.Result：The expression level of lncRNA ANRIL was significantly lower in RCC tissues（P 〈0.001）and cell lines（P〈 0.05）than paired normal tissues and 293 Tcells,and ANRIL was down-regulated in 34（72.34%）RCC tissues.The results indicated that the expression of ANRIL was associated with the TNM and AJCC stage of RCC（P〈 0.05）.However,no relationship was found between the expression of ANRIL and patients＇ age,sex and pathological types of RCC（P〉 0.05）.Conclusion：In RCC tissues and cell lines lnvRNA ANRIL is down-regulated,and this indicates that it may be involved in the tumorigenesis and development of RCC.
