摘 要:目的探讨集聚蛋白(Agrin)基因在人乳头瘤病毒(HPV)阳性和阴性宫颈癌细胞中表达的差异。方法以人乳头瘤病毒16(HPV16)阳性的宫颈癌Caski细胞和Si Ha细胞、HPV18阳性的宫颈癌He La细胞、HPV阴性的宫颈癌C33-A细胞为研究对象。通过Western blot和实时荧光定量聚合酶链反应(PCR)等方法分别检测Agrin mRNA和蛋白在不同宫颈癌细胞中的表达。结果实时荧光定量PCR和Western blot结果均显示,与HPV阴性的宫颈癌C33-A细胞比较,HPV16阳性的宫颈癌Caski和Si Ha细胞、HPV18阳性的宫颈癌He La细胞中的Agrin mRNA和蛋白表达均显著增加(P<0.01或0.05),Agrin mRNA和蛋白在4种宫颈癌细胞中表达的顺序为:C33-A细胞<Si Ha细胞<Caski细胞和He La细胞。结论 HPV阳性的宫颈癌细胞中Agrin基因的表达显著高于HPV阴性的宫颈癌细胞,为深入研究Agrin基因与宫颈癌的关系及相关机制奠定了基础。Objective To investigate the expression of Agrin gene in human papillomavirus( HPV)-positive and-negative cervical carcinoma cells. Methods Cervical carcinoma Caski cells( HPV16-positive),Si Ha cells( HPV16-positive),He La cells( HPV18-positive) and C33-A cells( HPV-negative) were taken as the research objects. Real-time fluorescence quantitative polymerase chain reaction( PCR) and Western blot methods were used to detect the expressions of Agrin mRNA and protein in different cervical carcinoma cells. Results Real-time fluorescence quantitative PCR and Western blot results indicated that compared with C33-A cells( HPV-negative),the Agrin mRNA and protein expressions in Caski cells( HPV16-positive),Si Ha cells( HPV16-positive) and He La cells( HPV18-positive) were significantly increased( P 0. 01 or 0. 05). The order of Agrin mRNA and protein expressions in the four kinds of cervical carcinoma cells was as follows: C33-A cells Si Ha cells Caski cells and He La cells. Conclusion The expression of Agrin gene in HPV-positive cervical carcinoma cells is significantly higher than that in HPV-negative cervical carcinoma cells,which has laid a foundation for further research on the relationship between Agrin gene and cervical carcinoma and its mechanisms.
